Assembly of the Adsys Controls MIRS system This document explains the assembly procedure of the Microscopic Imaging Research Station (MIRS) in a step by step manner from the individual components to a fully functional system. Instructions will be given for assembly and integration of the microscope assembly, microscope stage, the 150W light source, microscope motor controller, and the Adsys Controls ProLight Image Processing Module. This document is a compliment to the software user guide involving installation and operation of the Adsys Controls FPGA Image Processing Toolkit (FIPT), ProLight Image Processing Module, as well as the MIRS software reference design user's guide. II. Pre-Assembly Requirements Tools: Microscope Assembly: o 3/16 Hex Wrench (For M6 bolts) o 2mm Hex Wrench (For microscope set screws) o Micro Phillips Head Screwdriver Microscope Stage: o 5mm Hex Wrench (For M6 bolts) o 3mm Hex Wrench (For Post Clamp) o Flat Head Screwdriver (4mm wide head preferably) o 10mm Extended Socket Driver (included with system kit) Adsys Controls ProLight Image Processing Module: o Micro Phillips Head Screwdriver Power: 150W Light Source: o Standard NEMA 5-15 Socket 120VAC 60Hz Input Microscope Motor Controller: o Standard NEMA 5-15 Socket VAC 47-63Hz Input Hardware: Adsys Controls ProLight Image Processing Module: o PXI or PXIe/CompactPCI embedded controller on NI PXIe Chassis (MIRS software PXIe-796XR Series FPGA Module Currently Only Supported) MIRS Assembly Guide 4

5 IV. Assembly Instructions There will be five sets of sub-assembly installation guides involving the Microscope Assembly, Stage Assembly, Light Source, Motor Controller, and the ProLight image processing module. Please complete the instructions in the order they appear in the following text to ensure proper and safe installation. A. Microscope Stage This set of instructions will involve the steps needed to assemble the microscope stage which can be seen in the CAD rendered image below. The microscope stage consists of the Base Plate, Pedestal Pillar Posts, Z-axis Manual Positioner Stage, XY-axis Manual Positioner Stage, Optical Support Rod, Universal Mounting Plate, Modular Riser Plates, Post Clamp, Clamping Forks, and their hardware. 1. Procure the Base Plate from the packaging as well as the four pillar posts. Each pillar post will have a removable bolt attached to it. Using a flat head screwdriver (4mm wide head is optimal), back out the bolt to the top edge of the post such that half of the bolt is in the post and the other half out. MIRS Assembly Guide 5

6 2. Take each pillar post and attach them to the outermost corner thread-holes on the Base Plate. It may be needed to use the screwdriver to start the threading into the Base Plate. Lastly, tighten the pillar post to the Base Plate by hand to ensure the post will stay attached. (using loc-tite is optional). 3. Now obtain the handles from the packaging as well as four M6x16mm bolts. A 5mm Hex Wrench will also be needed for the M6 bolts. Take each Handle and line the holes up to the third thread-hole from the front and back on the Base Plate and attach it with the M6 bolts. The handles will be on the Left and Right edges of the Base Plate and centered from front to back. See images below. 4. Next, attach the Modular Riser Plate to the Base Plate. To attach the riser plate, two M6x25mm bolts will need to be obtained from the packaging. Position the riser plate such that the two countersink holes are facing upwards and in parallel to the right/left sides. Move the riser mounting holes such that the front hole matches up with the 8 th Base Plate hole from the front and the center hole from the left and right. See images below. MIRS Assembly Guide 6

7 5. We will now focus on adding the Universal Mounting Adapter, Z-Axis Manual Positioner Stage, and the XY-axis Manual Positioner Stage. First install the universal mounting adapter onto the Base Plate by positioning the center slot on the mounting adapter such that it is in line with the center column of Base Plate holes centered from left to right. Using two M6x12mm bolts, mount the plate such that the front edge of the mounting slot is pressed up against the front bolt, which is in the fourth Base Plate hole from the front. Also place the other bolt in the other available hole directly behind it. Be careful to watch that the adapter plate is relatively square to the Base Plate as it the bolts are tightened down. This ensures the stages will be correctly positioned under the microscope. See image below. MIRS Assembly Guide 7

8 6. Next will be the addition of the XY Manual Positioner stage. Obtain both the stage and four M6x6mm low profile bolts from the packaging. Position the stage such that the Y-axis Positioner (front to back travel) is on the bottom of the X-axis Positioner (left to right travel) with the micrometer on the side nearest the front. To begin installation, first turn the micrometer clockwise to its full extent to reveal the front mounting holes. Attach the stage to the universal mounting adapter as in the image below ensuring it is square before tightening. After the front bolts have been tightened, turn the Y-Axis micrometer counter-clockwise to its other extent to reveal the back mounting holes. Again use two M6-6mm low profile bolts in these mounting holes. See image below. 7. The Z-Axis Manual Positioner can now be attached on top of the XY stage. Obtain four M6x12mm from the packaging and position the Z stage on top of the XY stage such that the micrometer is on the side nearest to the front of the Base Plate. See image below. 9 8. Now that the stage is nearly complete, obtain the Optical Support rod and the 10mm extended socket driver from the packaging. Insert the socket driver down the center hole of the support rod and carefully position the support rod s bolt over the 1 st riser plate hole from the front and center hole from the left/right. Use the socket driver to attach the support rod to the riser plate while making sure to align the teeth on the back of the support rod such that they are pointed towards the back of the assembly. See images below. 9. Place the Slip-On Post Clamp onto the support rod using the 3mm Hex wrench at a distance around 100mm from the top (will be adjusted again later in assembly). See image below. 10 B. Microscope Assembly This set of instructions will involve the steps needed to assemble the microscope assembly, which can be seen in the CAD rendered image below. MIRS Assembly Guide 10

11 This assembly will include the 12X Motorized Microscope Body, 2X Body Tube Adapter, C-mount Coupler, High Speed Imaging Camera (Plus Power Supply), Microscope to Stage Mounting Plate, Rod Clamp, Microscope Objective Coupler, an objective (20X or 50X), and the optional filter Accessories. 1. To begin assembly of the microscope sub-assembly, obtain the 12X Motorized Microscope Body and 2X Body Tube Adapter from the packaging. Be very careful to not drop or bump these components as there are very sensitive optical components internally. First remove the dust covers from both components, which are used to protect the optics from dust particles. To attach these two components, carefully place the 2X Body Tube atop the 12X Motorize Microscope Body such that the Flange on the Microscope body goes inside the opening on the 2X body tube that does not have a similar Flange. To secure these components, use a 2mm Hex Wrench to tighten the 3 set screws around the perimeter of the 2X Body Tube. It is VERY IMPORTANT to tighten these screws equally as this aligns the Optical path in the microscope up to the sensor within the camera. See image below. 2. The next step will be to attach the C-mount Coupler to the flange on the top of the 2X adapter tube. Similarly to the step above, use a 2mm Hex Wrench to tighten the three set screws around the perimeter of the C-mount Coupler. Again, it is VERY IMPORTANT to tighten these screws equally as this aligns the Optical path in the microscope up to the sensor within the camera. Refer to the image below. 3. Next will be the addition of the Camera to the C-mount Coupler. To do this, remove the camera sensor protection cap and then quickly position it atop the C-mount coupler and screw it in by rotating it clockwise. The necessity for a quick transfer is to ensure dust particles do not get into the MIRS Assembly Guide 11

12 opening where the sensor is. The image quality could be affected as dust particles settle onto the sensor. See image below. NOTE: After installing the microscope assembly to the full system, the C-mount Coupler may need to be adjusted. If so, re-adjust the positioning of the C-mount Coupler such that the bottom of the camera (side closest to the power plug) is facing towards the front of the Base Plate and square. This will allow the image back to the user to be displayed in a logical manner. 4. Carefully set aside the assembly thus far in a location where it will not be bumped or fall. Next procure the Rod Clamp and the Microscope to Stage Mounting Plate as well as two M6x16mm bolts and two M6x25mm bolts. First attach the Mounting Plate to the Rod Clamp to match the following image. The center two bolts will be M6x16mm and the two bolts on the left and right will be M6x25mm. 5. Next will be the installation of the Mounting Plate/Rod Clamp assembly from the previous step to the microscope assembly from step 3. Four of the 4-40 bolts are needed which require the use of a small Phillips or standard flat head screwdriver. To install the mounting plate assembly to the microscope first locate the four small bolt holes in a square pattern on the opposite side of the motors on the 12x Microscope Body. Next, place two of the 4-40 bolts into the thru holes on the mounting plate and carefully position it over the holes on the microscope. Take extreme care not to scratch the outer body of the microscope as the mounting plate is placed onto the microscope. MIRS Assembly Guide 12

13 Tighten the two 4-40 bolts securely (hand tighten and do not over-tighten as the threads on the microscope can strip out). Next do the same for the final two bolts. Refer to the image below. 6. Lastly will be the installation of the Objective and the Microscope Objective Coupler. NOTE: This assembly can be done after the current Microscope Assembly is attached to the Stage Assembly near the end of Section C of this assembly guide to ensure the objective will not be damaged in transfer. To install the microscope objective to the Microscope Assembly, obtain either a 20X or 50X Objective from the packaging, as well as the Microscope Objective Coupler. Attach the coupler to the Objective and then carefully thread the objective into the lower opening of 12X Microscope Body. Use extreme caution in this installation and use both hands to ensure it does not drop or fall. As a safety precaution, place a padding of some kind underneath the objective in the slight chance it is dropped. See the image below. 7. [OPTIONAL] If there is a desire to use the filter accessories included with the system please follow the following instructions. To place a filter in the optical path of the microscope, obtain the filter MIRS Assembly Guide 13

14 holder from the packaging. Remove the knurled ring from the top of the filter holder which opens up the filter holding area in the filter holder. There is an adjustable ring in this area as well that can be adjusted to fit different thickness filters. To install the filter carefully place it in this area (careful to not leave finger prints) and then screw the knurled ring on top of the filter back on top of it to hold it in position. Adjustment of the ring may need to be done multiple times to ensure the filter is held tightly, but not too tight. Next attach the filter holder to the objective of choice and then insert into the microscope similarly to step 6. C. Microscope Assembly to Stage Assembly This short set of instructions will guide the installation of the Microscope Assembly form section B to the Stage Assembly from section A. 1. First, carefully obtain the microscope assembly from the position it was resting in after section B. Be sure to use two hands and grab the microscope by the main 12X body tube to ensure the alignment of other components is not compromised. Carefully position the Rod Clamp on the Microscope Assembly onto the Optical Support Rod on the Stage Assembly. Ensure the slot on the Rod Clamp matches up with the gear teeth on the Optical support rod. Slowly move the Microscope Assembly down on to the Post Clamp as a rest place. Next, be sure to tighten the handles on the rod clamp to clamp the microscope in place. This ensures the microscope assembly will not move. Refer to the image below. NOTE: Steps 6/7 of section B may be followed if not done so already. MIRS Assembly Guide 14

15 D. 150W Light Source This set of instructions will involve the steps needed to connect the light source to the microscope assembly. The contents of this installation involve the 150w DC regulated Fiber Optic Power Supply, Flexible Light Pipe for Coaxial (3 foot length), 8mm Diameter Fiber Input for 12X body Tube, and Standard C13 Plug AC Power Cord (5 foot length). In the image below, it can be seen that the light source has a port on its front which takes the input of the fiber optic light pipe, as well as a dial for adjusting the intensity of light. 1. To install the light source to the microscope assembly, obtain the 8mm Diameter Fiber Input for 12X Body Tube from the packaging as well as the 3 foot Flexible Light Pipe. Remove the protective caps from the Light Pipe and be very careful not to scratch the ends as this will affect the lighting quality. Firstly, the Fiber Input Adapter will need to be attached onto the microscope by screwing it into the port on the microscope similarly to the image below. MIRS Assembly Guide 15

16 2. In the above image, it can also be seen how the end of the Flexible Light Pipe will fit into the end of the Fiber Input Adapter on the microscope. Use the thumb screw to tighten the Light Pipe to the adapter. Take the other end of the light pipe and insert it into the port of the 150W Light Source and tighten that thumb screw as well. 3. Lastly, Take the AC power cord and insert it into the back of the 150W Light Source and plug it into a 120VAC power outlet. The on/off switch is on the front of the Light Source directly between the Light Intensity knob and the Light Pipe Port. E. Microscope Motor Controller This set of instructions will involve the steps needed to connect the serial cables coming from the microscope assembly to the Motor Controller. The contents of this assembly include the Motor Driver Module (plus AC Adapter) and an RS-232 Serial Cable. The image below shows the installation of each external component mentioned in the previous steps. 1. The first step to installing the Motor controller to the microscope assembly is to find a proper location for the motor controller, which may be on top of the 150W Light Source in a location near the microscope. Route the serial cables coming from the motors on the microscope towards the back of the motor controller which has two serial ports labeled axis 1 and axis 2. Axis 2 corresponds to the Focus Motor which is located at the bottom of the 12X Microscope Body, leaving Axis 1 as the Zoom Motor near the top. Wire each line accordingly to the back of the motor controller. MIRS Assembly Guide 16

17 2. Next, obtain the AC power adapter for the motor controller from the packaging as well as the RS- 232 Serial Cable. Attach the power adapter to the motor controller as well as the serial cable in the serial port on the back of the controller. NOTE: Before turning on the motor controller with the toggle switch in the back, ensure that there is nothing directly under the objective, as the objective will move up and down as the motors initialize after power-up. NOTE: Serial is only supported in software currently even though there is a USB physical connection. A USB-to-Serial conversion cable can be used if necessary to run the software if a RS-232 port is not available on the controller. F. Adsys Controls ProLight Adapter Module This section describes the installation of the Adsys Controls ProLight adapter module. This requires the LabVIEW FPGA module to already be installed in the chassis. The Adsys Controls ProLight 1101 adapter module is shipped in antistatic packaging. Standard precautions for ESD safety should be observed. The adapter module should be installed with the chassis powered down. 1. Gently insert the guide pin on the Adsys Controls ProLight 1101 adapter module into the corresponding connectors on the NI FlexRIO board already installed in the PXI chassis. With a possible tight fit, do not force the adapter module into place. MIRS Assembly Guide 17

18 2. Tighten the captive screws on the Adsys Controls ProLight 1101 adapter module to the FlexRIO front panel. 3. The following diagram shows the two connection points for Camera Link Base and Camera Link Full connectors. These connectors are Mini-Camera Link connectors (SDR). Obtain the Camera Link Cables from the packaging and install them to the ProLight module. It is recommended to label each cable so that there is no confusion as to which cable is used for which line. Next take the cable running from the Base port on the ProLight and route it to the top of the microscope to connect to the port on the camera that has a small B labeled on it. Similarly, do the same for the cable connected to the Full port. Camera Link Base Connector Camera Link Full Connector Digital IO Connector 4. The standard Digital IO connector is shown below with details. This connector will not be necessary for MIRS operation but is placed here for reference.
