The platform should be placed conveniently as to minimize the need to move after initial setup. The area/room the platform is placed in should have consistent, even lighting as to prevent errors due to fluctuations in ambient lighting. Standard lighting not to exceed 675 lux. SETTING UP THE PLATFORM Place the microscope in the area selected for use. Loosen the three screws on the Cradle that attach the cradle to the C-Mount Adapter. Remove the two screws from the Cradle Support. Attach the Cradle Support to the Cradle with the two screws you removed. Remove the two screws located on the top of the back of the Microscope. Remove the C-Mount Adapter Cap and align the Cradle with the tube of the adapter sliding them together. (Note: the screw holes on the bottom of the Cradle Support should line up with the holes you just opened in the previous step) Attach the Cradle Support to the Microscope using the two screws you removed. Balancing The Cradle Install the application Bubble Level from the App Store if not already on your iphone. Open the application and switch it to Surface Level mode by swiping the app screen to the left. Place the phone on the cradle over the main body of the platform. To adjust back-to-front you can loosen the thumbscrew on the adapter between the cradle and the c-mount of the platform, then adjust the adapter by extending or retracting it until the bubble is center. To adjust left-to-right use the hex key to tight or loosen the screws holding the cradle to the adapter until the bubble appears center. 3

INITIAL CALIBRATION Centering The Condenser With no sample on the stage, slowly close the field iris by turning the dial around it clockwise. As the edges of the iris come into the field of view, adjust the center of the condenser using the thumbscrews located on the front left and front right sides of the condenser. When complete, the hexagon of the field iris should appear in the center of your field of view. Begin turning the dial on the field iris counterclockwise until the edges of hexagon are just outside the field of view. Aligning The Phase Rings Note: The phase rings should be aligned upon receipt of the unit, do not adjust the screws on the ring holder in the condenser unless you ve verified misalignment. Remove one ocular lens and replace with the telescopic ocular provided. Loosen the thumbscrew on the telescopic ocular and begin slowly pulling the telescope outward. Once you can see the light and dark rings clearly, tighten the thumbscrew to hold the position of the telescope. (DO NOT OVER-TIGHTEN) If the light ring does not appear to sit neatly inside the dark ring, note the position of the two rings. Remove the phase ring holder from the condenser. Carefully adjust the position of the phase ring using the small hex key (allen wrench) provided on the screws located on the back right and front right edges of the ring holder using 1/4-3/4 turns for each adjustment. Return the holder to it s place inside the condenser, be sure that the notch for locking into the condenser is facing away from you and the handle of the ring holder is located on the right side of the condenser. Check the position of the rings through the telescopic ocular, if they are still out of alignment repeat steps 5-7 and check again until they are aligned. Once finished, return the telescopic ocular to its initial position and replace with the standard ocular. MAINTENANCE AND BEST PRACTICES Looking After the Microscope: - All standard measures of protection/cleaning of the optics of the microscope apply. Normal (laboratory) usage should not require cleaning of any of the parts of the microscope. In case something goes wrong or you are not sure, please contact technical support. - The eyepiece should not be exposed to dust. This may however happen. If it does, you will notice that a permanent speckle is visible on all of your images. First, check that the iphone lens to confirm it is clean. If this is the case, dust may be on the eyepiece itself. In this case, we recommend that air is blown (see FAQ) to clean the eyepiece. - When not being used, the microscope should be covered by a plastic sheet. - The microscope has a battery and is able to function for at least 8 hours without interruption. The iphone can also be charged from the socket at the back of the microscope. When a power socket is available, and the iphone is charged, we recommend to have both microscope plugged in. Note: the iphone cannot charge at the same time as the microscope battery. Looking After the iphone - The iphone should be cleaned thoroughly and regularly with a clean cloth. The user should take particular care to ensure that the camera lens and screen is free of obstruction and smudges. - It is imperative that the iphone is protected from scratches to the screen and the lens of the camera. - It is important to make sure the iphone and the xr Fiber app are up to date. The user can ensure that the iphone is up to date by tapping the Settings icon on the home screen and navigating through General - Software Update. - You will be notified by email and message whenever an update is needed. In case this update is critical our Support will call you to make sure proper steps are taken to this effect. 4 5

II. SOFTWARE: GETTING STARTED WITH XR FIBER HARDWARE: SETUP OF XR FIBER PLATFORM STARTING CONDITIONS xr Fiber has been installed on your iphone xr Fiber Platform has been set up in a location selected based on the parameters given in Placement of the xr Fiber Platform Your iphone and xr Fiber Platform are sufficiently charged or plugged into a standard AC outlet REGULAR CALIBRATION At the start of the day, before any samples have been analyzed, the condenser and phase ring alignment should be checked. Periodically an HSE/NPL slide (not included) should be loaded onto the platform and a user should verify that blocks 1-3 are keenly visible, blocks 4-5 are at least partially visible and blocks 6-7 are not visible (per NIOSH 7400 guidelines). If you suspect visibility issues, the cradle balance should be checked. SAMPLE PREPARATION Equipment and reagents for sample preparation are the same as outlined in the NIOSH 7400 guidelines with differences as follows: - The labelled slide being used should be loaded onto the Slide Preparation Template included with your xr Fiber Platform. - The quarter filter, after being cut, should be placed with the tip over the hole and the curve following the semi-circle outline on the template. Note: Sample quality can effect performance greatly so care should be taken to fully dissolve the filter and bubbles should be avoided as much as possible when placing the cover slip. RUNNING XR FIBER After turning on the xr Fiber Platform, load the iphone onto the Cradle and open the xr Fiber app. If this is the first time using the app: - Press the Settings button. - Enter the user s name and company details under the User tab. Under the Diagnostic tab you can: - Select the type of analysis to be performed - The frequency of image purging (Never, Monthly, Bi-monthly) Printers can be installed under the Print tab. You can also select the type of paper to be used, number of copies and color vs. monochrome (B&W) formats. After you ve completed all of the following steps, select Save in the bottom right corner of the screen. - Once you are ready to run your first test, press the Start Measure button in the center of the home screen. - A menu window should appear with the microscope s serial number listed. - Select the scope by clicking on the serial number (to make connections easier in the future, the user should toggle the switch next to remember my microscope ) and pressing the Connect button. - You should see a message window saying Connected to Microscope confirming the phone has connected to the platform. Note: Once a microscope has been saved, if you wish to connect to a different scope, you can select the Disconnect button in the top right of the testing screen. There may be a delay before the microscope selection menu appears again. - Place your sample on the stage, in the stage clip, with the tip of the filter pointing to the user s left, make sure that the sample is completely flat against the stage and held firmly. Note: Ensure that due care is taken when inserting the slide into the Microscope. The clip must be returned to its natural position with care in order to house the slide properly. - The randomly generated sample number at the top of the screen can be changed by the user. Simply tap on the number and the keyboard controls will appear for deleting and replacing the sample number. 6 7

There are two options for positioning the sample: Manually Positioning - Select the Manual button in the corner of the viewing window, this opens the manual controls for the microscope. - Using the directional pad on the right side, move the stage so the corner of filter is under the objective. - Using either the left + - pad or the focus knob, bring the sample into focus in the viewing window. - Close the manual controls by pressing the Auto button that replaced the Manual one. Automatic Positioning - Press the Zero button in the lower right corner of the viewing window. - After the analysis is complete, you will be brought to the Results screen where you can receive your results in fibers/fields, fibers/mm 2 and fibers/cc. Note: When you select the fibers/cc tab you will be prompted to enter the sample volume in order to receive results. The default value is 1200L and can be used by selecting the Keep Default button. - Images can be reviewed, exported or printed by pressing the Images Saved button. Note: Images can be sorted by All, Positives (fibers present), Not valid (discarded fields) and Negatives (no fibers present). Note: Automatic Positioning requires care in sample preparation and use of the Sample Preparation Template. - Using the focus knob, bring the sample into focus in the viewing window. - Once the sample is in position and in focus, press the Start Diagnostic button in the center of the bottom of the screen to begin the analysis. Note: When the sample appears inconsistent, a pop up window will notify you, after which you have two options: 1) Bring the field into good focus with the focus knob and continue (Recommended if the user feels the sample is consistent and the notification is in error). 2) Abort the sample and read it manually (Recommended if the user feels the sample is truly inconsistent). Note: When a sample appears overloaded, a pop up window will notify you, after which you have two options: 1) Bring the field into good focus with the focus knob and continue (Recommended if the user feels the sample is not overloaded and wishes to continue the analysis). 2) Abort the sample and report it as Overloaded. QUALITY CONTROL There are three settings available for how you want the xr Fiber app to handle Quality Control (QC): - None - Not recommended for use with the NIOSH 7400 Guidelines - Automatic Selection - At the end of the day, the user opens the Test Log (found at the bottom of the home screen). 1/10 samples (10%) run that day will be selected at random to be analyzed again. Recommended for users without another form of QC selection system. - Manual Selection - This option allows users to slide a sample number from left-to-right to select a sample to be run again (at no cost) for QC. Recommended for users with an existing QC sample selection system (i.e. LIMS). If a user would like to review the sample information and results for previous samples, press the Tests Log button at the bottom of the home screen. Samples are sorted by date the test was run. 8 9
