GeneMapper TM ID-X Software supports a multi-user database configuration and provides a set of streamlined data analysis and sample review tools. The software offers a comprehensive quality value system that can be optimized and validated for use in both expert system and traditional data analysis workflows. The software supports genotyping applications on the ABI PRISMTM 310, 3100, 3100-Avant, Applied Biosystems TM 3130, 3130xl, 3500, 3500xl Genetic Analyzers and Applied Biosystems TM 3730 DNA Analyzer. After reading the updates included in this release, each laboratory should decide whether internal validation testing is necessary for its system to comply with laboratory guidelines or regulations. 1
DESCRIPTION OF GeneMapper TM ID-X Software V1.5.2 REQUIREMENT Problem Statement: In GeneMapper ID-X Software v1.5, after printing a sample plot user was unable to see the gray color bins on sample plot due to lighter shade of the bins. When the sample type was set as Negative Control the sample was assigned a red CGQ flag due to failed CC flag. This was because, the presence of IQC peaks trigger the Allele Number Rule of negative control (no peaks are expected in a valid negative control). Solution: Changes made in GeneMapper TM ID-X Software v1.5.2 include: Change in the gray color shade to add more contrast such that upon printing of sample plot user will now be able to see the gray color bins. Enhancement to the Allele Number Rule to not raise CC flag if the observed IQC allele value is present in the IQC marker range for Negative Control sample type. The valid allele values for marker (IQCL and IQCS) can be defined in the QualitySensor.properties file (with their respective kit name and panel name). These values are user editable. Details on how to edit this file is described in the later section of the document. SYSTEM REQUIREMENTS The minimum supported computer hardware and system requirements for GeneMapper TM ID- X Software v1.5.2 is identical to GeneMapper TM ID-X Software v1.5, and so are the OS options supported. UPGRADE TO GeneMapper TM ID-X Software V1.5.2 Pre-requisites: GeneMapper TM ID-X Software v1.5/1.5.1 is installed successfully for both full and client version. No GeneMapper TM ID-X process is running. If GeneMapper TM ID-X is already installed on the machine; reboot the machine to ensure that GeneMapper TM ID-X process is not running. Back up any critical data User upgrading GeneMapper needs to have admin privileges. 2

b. Installer should display the Welcome window as shown below. Click Next to continue. 4
c. Install should display the "Ready to install" window as shown below. Click Install to continue. 5
d. The installation in progress and setup status window should get displayed as follows: 6
e. Click Finish to finish the installation. f. Upgrade to GeneMapper TM ID-X v1.5.2 base is complete. 2. Repeat above step (#1) for each full and client install on the same network. 7
3. Please refer following log files for additional information: Gmidx_15_bin_color_patch_logs.log file at location <GMIDX_installed_Dir>\GeneMapperID-X folder. VERIFICATION OF INSTALLED VERSION (Ensure the patch version 1.5.2 is applied) 1. Go to Control panel ->All Control Panel items->programs and Features. 2. Check the version string for GeneMapper TM ID-X software (It should be GeneMapper ID-X v1.5.2 as shown below). 3. Check the existence of the release note at the <install location>\ Docs folder STEPS FOR EDITING QualitySensor.properties FILE 1. Upon successful installation, the property file can be found under <installed location>\appliedbiosystems\genemapperid-x\ QualitySensor.properties 2. To make changes to the file: a. Open the file in a notepad or a text editor. b. Make necessary updates (maintain the format and hierarchy). c. Save the file at the same location with the same name (overwrite the original file). d. Restart GeneMapper TM ID-X. 8
KNOWN ISSUES AND LIMITATIONS Gray color shade may get affected because of screen brightness; contrast setting s and angle of laptop screen. Please try to adjust these things if you don t see bin color properly. Any other known and previously documented issues with GeneMapper TM ID-X Software v1.5 will not be addressed in this software release. 9
