Standardizing Meter and ph Electrode ph measurement is a systemic measurement process that includes voltage measurement (the ph meter), a sensing device (ph electrodes) and known standards (ph Buffers). As ph electrodes vary in their response, it s important to standardize your ph meter and electrode to compensate for electrode variation. The more often you standardize your meter and electrode the more accurate your measurement. In addition, accurate ph measurement is dependent on standardizing the meter and electrode with the appropriate ph Buffers. The ph-11 Meter offers a choice of fifteen ph buffers in three buffers sets. The most common buffer set (North America) includes ph Buffers 4.01, 7.00, The default buffer set for the ph-11 meter includes ph Buffers 4.01, 7.00, and As the performance of ph electrodes is non-linear (typically), we strongly suggest a Three Point Standardization. If you are unable to perform a three point standardization, perform a Two Point Standardization that brackets the ph value of your sample. ph Buffers are a critical component in accurate ph measurements and they have a life expectancy. To warrant that you have properly standardized your ph meter and electrode, check the date code on your ph buffers to ensure they have not expired. Use fresh buffers each time you standardize your meter and electrode. Now your ready to standardize your Meter and ph Electrode: 1) After you have conditioned your electrode, rinse it with distilled or deionized water, blot dry (do not wipe electrode). Immerse electrode* in ph Buffer 7, provide moderate stirring. 2) Press CAL Key, CAL 1 appears in top display and. When the stability icon appears, Press CAL Key again. The display will flash 7.00 ph for a few seconds until the meter has detected a stable reading at which time END will appear (flash) in the display followed by 7.00 ph indicating you have standardized the meter and electrode at ph Buffer 7. CAL 2 appears in the top display prompting you to perform a second standardization (Two Point). 3) Rinse electrode (see #1) and immerse it in ph Buffer 4, provide moderate stirring. When the stability icon appears, Press CAL Key. The display will flash 4.00 ph for a few seconds until the meter has detected a stable reading at which time END will appear (flash) in the display followed by 4.00 ph indicating you have standardized the meter and electrode at ph Buffer 4. CAL 3 appears in the top display prompting you to perform a third standardization (Three Point - Recommended). 4) Rinse electrode (see #1) and immerse it in ph Buffer 10, provide moderate stirring. The display will flash ph for a few seconds until the meter has detected a stable reading at which time END will appear (flash) in the display followed by ph indicating you have standardized the meter and electrode at ph Buffer 10. 5) The meter will return to the measurement model and the L (Low), M (Medium) and H (High) icons will appear in the bottom left hand corner of the display indicating you have performed a three-point standardization. *If you choose to perform a One or Two Point standardization, Press Enter Key at the end of Step 1 or Step 2 listed above. If you perform a One Point Standardization the M (Medium) Icon will appear in the display. If you perform a Two Point Standardization the L (Low) and H (high) Icons will appear in the display. When performing a Three Point Standardization the L (Low), M (Medium) and H (High) Icons will appear in the display. Page 5

ph is temperature dependent, see page 7. The GeneMate meters have two temperature compensation options; Manual Temperature Compensation (MTC) or Automatic Temperature Compensation (ATC). To ensure the most accurate measurement we recommend the use of the Automatic Temperature Compensation feature. If you purchased a ph-11 Meter kit that did not include the ATC Probe (Part # ATC-01) you can purchase this item separately. However, the Manual Temperature Compensation feature on the GeneMate Series Meters (ph-11 models) can be adjusted to match the temperature of the ph buffers during the standardization process or when measuring samples. If you chose to utilize the Manual Temperature Compensation function, see pages 7 & 10 for additional instructions. When standardizing your meter OR measuring a solution using Automatic Temperature Compensation (ATC) please immerse the ATC probe and Electrode completely into each buffer or solution to ensure the actual temperature of the buffer or used is used in the calculation process. Standardizing Meter and OPR (Oxygen Reduction Potential) Electrode ORP or redox potential is a millivolt measurement that indicates the oxidizing or reducing capacity of a solution. ORP values are used to control or monitor samples that require a specific amount of oxidants or reductants. To standardize your Meter and OPR Electrode: 1) Clear out any Calibration (Standardization) Points that may be stored in memory 2) Set the mode of measurement on your meter to ORP Mode, see page 4. 3) Install ORP Electrode, see page 4. 4) Program an appropriate Measurement Interval Setting that is appropriate for you sample, page 7, P2. 5) Immerse the probe in to the ORP Standard and Press Cal Key. When the instrument detects a stable reading the absolute mv value of the standard becomes zero relative millivolts and the meter will display O in the ORP mode. 10 In addition to optional temperature settings (C of F), GeneMate Meters also have Automatic Temperature Compensation (ATC) and Manual Temperature Compensation (MTC) functions. Automatic Temperature Compensation is the recommend method as the temperature of the ph buffers and your samples may change from one to the other or even during the measurement process. If you did not purchase a meter kit that included the ATC Probe you can purchase one separately. If you use the MTC option simply PRESS the Up / Down arrow keys to adjust the temperature setting. Please note that programming a temperature into the meter that does not match the true temperature value of your samples will adversely effect the accuracy of your measurement. If you do not adjust the temperature while operating in the MTC mode the meter assumes the temperature of the sample is 25C and calculates the ph reading based on this temperature value. ATC is the best method to ensure your meter is measuring temperature and therefore ph, accurately. 12 ph Theory ph is the measurement of the acidity or alkalinity of a solution, calculated by the relative number of hydrogen ions (H+) or hydroxyl ions (OH-) present in a solution. Acidic solutions have a higher relative number of hydrogen ions, while alkaline (basic) solutions have a higher relative number of hydroxyl ions. Acids either dissociate (split apart) to release hydrogen ions or react with water to form hydrogen ions. Bases are substances that dissociate to release hydroxyl ions or react with water to form hydroxyl ions. ph is defined as the negative logarithm of the hydrogen ion activity: ph = - log [H+] At 25 C, a reading of 7.0 ph is neutral, solutions with a ph value less than 7 are acidic and solutions with ph value above are alkaline. The typical ph range is 0 to 14, although solutions containing non-water solvents can have ph values outside this range. ph is also logarithmic with each ph unit has ten (10) times as many hydrogen ions and the unit above it. At ph 3 there are ten (10) times as many hydrogen ions present in the solution that at ph 4 and one hundred (100) times more than at ph 5. ph measurement is based on the use of a ph sensitive electrode (typically glass), reference electrode, and a temperature probe to provide a temperature signal to the ph meter. Most electrodes available in today s market are combination electrodes a combination of the measurement electrode and reference electrode. Also available are Combination ph/atc (or 3-in-1) Electrodes which combines the measurement electrode, reference electrode and automatic temperature compensation probe (ATC). ph Values (typical) of Common Substances: ACIDS Hydrochloric Acid (HCl) 0.0 Lemon Juice 2.2 Vinegar 3.0 Wine & Beer 4.0 Coffee (Black) 5.0 Milk 6.6 NEUTRAL Pure Water 7.0 BASES Blood 7.4 Baking Soda (Sodium Bicarbonate) 8.5 Milk of Magnesia 10.6 Ammonia 11.0 Lye 13.0 Sodium Hydroxide (NaOH) 14.0 ph electrodes use a specially formulated, ph sensitive glass when in contact with a solution develops a voltage potential proportional to the ph of the solution. The reference electrode is designed to maintain a constant potential at any given temperature, and serves to complete the ph measuring circuit within the solution. It provides a known reference potential for the ph electrode. The difference in the potentials of the ph and reference electrodes provides a millivolt signal proportional to the ph value. ph electrodes are designed to produce a 0 mv signal at 7.0 ph, with a slope (theoretical) of mv per one unit of ph at 25 C. Page 10

ph and Temperature Compensation ph and temperature compensation is described by the Nernst equation, with the following equation: Temperature change affects ph Measurement in following two ways: 1) ph Buffer Values The value of ph Buffers change with temperature changes. Temperature and corresponding ph Values are typically found on the label of the ph Buffer bottle. The chart below provides data on US Standard Buffers. Please check the label on the buffers used when standardizing (calibrating) the meter and electrode. U.S Standard Buffers Temperature (C) ph Buffer 4.00 ph Buffer 7.00 ph Buffer ) Electrode Efficiency In addition to the value of ph Buffers, the efficiency of the ph electrode is affected by temperature change. For hydrogen ion activity (ph) the Nernst factor is mv for every one unit of ph (ten-fold change in activity) at 25 C. This means that for every ph unit change, the total potential will change mv. However, the Nernst factor will change when temperature changes as temperature (T) is not constant. At 25 C the slope of the ph electrode is mv/ph unit. At 0 C the slope value is approximately 54 mv/ph, and at 100 C the slope value is approximately 74 mv/ph. The millivolt output of the glass ph electrode changes with temperature in accordance with the Nernst equation. As the temperature increases, the millivolt output and the slope of the electrode changes accordingly. Page 11

Electrode Care & Maintenance Before using your ph meter and electrode; standardizing (calibrating) or measuring: Soak the electrode for one hour in an electrode filling solution, KCL solution or electrode storage solution. ph electrodes vary in composition, please reference the operation manual on the electrode you are using for filling, storage and maintenance instructions. Open the fill hole before standardizing or measuring. Close the fill hole when the electrode is in storage. DO NOT let the internal filling solution evaporate completely. You might be able to rejuvenate the electrode when it dies out, but most likely this will ruin your electrode. Risen the electrode with distilled or DI (deionized) water between ph buffers when standardizing and between measurements. Blot the electrode dry, do not rub or wipe the electrode. Re-standardize the electrode and meter on a regular basis. If you use the meter daily standardize every 30 to 40 measurements or a least once per day. If you go a long period (more than a day) between measurements, re-standardize. Always use fresh buffers when standardizing. Always perform a three-point standardization (calibration), i.e.: ph Buffer 4, 7 and 10. Proper electrode care and maintenance is essential for accurate ph measurement and will extend the life of your electrode. Troubleshooting ph Electrodes All GeneMate Series Meters come with a millivolt (mv) mode. The millivolt mode can be used to check the performance of your ph electrode. To test the performance of your electrode*: 1) Clear out all ph Buffers and Relative mv Offset that are currently stored in memory. To clear buffers and relative mv offset, you will need to reset the meter to the Factory Default Setting, see page 7, P6. Please note that you will need to re-program settings P1 through P56 anytime you utilize the Factory Default Setting. 2) Press the Mode key and Toggle to the mv Mode 3) Prepare fresh samples of ph Buffer 4, ph Buffer 7 and ph Buffer 10. (Make sure the buffers have not surpassed their expiration date) 4) Rinse the electrode with DI Water and place into ph Buffer 7: Value should equal 0 mv +/- 30mV to 60mV (Depending on Electrode). 5) Rinse the electrode with DI Water and place into ph Buffer 4: Value should be 160 mv HIGHER that the Value of ph Buffer 7 6) Rinse the electrode with DI Water and place it into ph Buffer 10: Value should be 160 mv LOWER that the Value of ph Buffer 7 (record reading). * Please note that this is a general test for the performance of ph Electrodes. Please refer to the operation manual of your ph Electrode for the specifications and mv tolerances and troubleshooting procedure. Page 12

Troubleshooting the GeneMate GpH Meter In addition to checking the performance of ph Electrodes the millivolt mode can be used to perform a quick check of your ph meter as well.
