6 01 Introduction 01 Introduction Thank you for purchasing an AmpliSpeed slide cycler. The AmpliSpeed offers superior thermal cycling performance and unique flexibility on either one (ASC100D), two (ASC200D) or four (ASC400D) independent slide units. With the ASC100E, ASC200E or ASC300E unit various upgrade possibilities are delivered. Controlling and definition of thermal cycling profiles is obtained with the new 640 x 480 pixel VGA touch screen display using an innovative graphical user interface for benchtop programming. 8 02 Safety Precautions 02 Safety Precautions! Attention! Please read safety precautions carefully! Do not allow any uninstructed personnel to operate the AmpliSpeed slide cycler! Please read the entire manual carefully before using the AmpliSpeed slide cycler as it contains all the important safety information. Do not allow any uninstructed personnel to operate the AmpliSpeed slide cycler. If the instrument is transferred to a third person please include this product manual. Warning: Surfaces of the AmpliSpeed slide cycler may get extremely hot, reaching temperatures of up to 99 C. This might cause serious burns or injuries. Please do not touch any surfaces of the instrument while running. Always close the lid to prevent people from touching the heated plate. Do not put materials on the heating plate that are not certified temperature stable. Warning: Before electrically connecting the device, please compare the specifications of the power outlet with the voltage requirements on the back of the instrument. Warning: Do not block the air intakes of the AmpliSpeed slide cycler, as this will dramatically reduce thermal cycling performance. Also overheating of the instrument may result in severe damage of the electronic parts inside. When placing more instruments next to each other please provide proper air circulation. Do not use the AmpliSpeed slide cycler in closed housings. Only integrated solutions of the AmpliSpeed instrument e.g. for the use in liquid handling platforms have especially been designed to work in low air movement environments. Clean air intakes of the AmpliSpeed on a regular base to prevent accumulation of dust particles that will inhibit proper air flow. Warning: Please ensure that no liquids are spilled on the heating plate and / or are entering the instrument. 10 03 Before you start 03 Before you start Unpacking the AmpliSpeed Place the shipping box with the arrows up, indicating the upper side. Open the sealing tape without damaging the parts inside. Do not use knifes, cutters or any other sharp objects to remove the sealing. Do not carry the AmpliSpeed slide cycler at the white display unit. Grasp the instrument on both sides of the silver-grey slide units and lift carefully. Remove the adhesive strips from the lid and the protective foil from the display unit. Please keep the original packaging material in case you have to return the instrument. 12 04 Setting up the AmpliSpeed 04 Setting up the AmpliSpeed Basic setup The AmpliSpeed system requires only minimum assembly. Place the instrument on a vibration free and flat surface. For the ASC100 / 200D plug in the power cord (see fig. 3). For the ASC400D connect the two delivered parts by stacking the units and tighten the selfcutting screws with a TX8 torx key (see fig. 8). Then plug in the power cord. Minimum space requirements for the ASC100D are 29 cm x 16 cm x 20 cm (DxWxH), for the ASC200D 35 cm x 16 cm x 20 cm (DxWxH) and for the ASC400D 48 cm x 16 cm x 20 cm (DxWxH). Upgrading the system For upgrading to higher configurations please switch off the system, disconnect the AmpliSpeed slide cycler from the power socket and remove the power supply by unscrewing the torx screws. Carefully pull the slide unit backwards without canting. Then stack the new slide units and connect the power supply again. Please do not use electrical tools like cordless screwdrivers and tighten the screws carefully not to strip the thread. Removing an AmpliSpeed unit The AmpliSpeed slide cycler is a modular system. Even if one slide unit is broken or has to be sent in for service or maintenance the others will remain fully functional. Minimum configuration of the stand-alone instrument is one display unit, one slide unit and a power supply. Please switch off the system first. Removal of either the slide unit or the control display when powered may result in severe damage of the system. After switching off the system please wait for 10 seconds and loose the torx screws at the sides of the unit with a torx TX8 tool (see fig. 8). Carefully pull the slide unit backwards without canting. Connect the remaining units again and tighten the screws. Please be careful not to strip the thread.! Attention! Do not connect or disconnect an AmpliSpeed slide unit when the system is switched on. Irreversible damage of the system may occur. Connecting the power supply The AmpliSpeed comes with country specific power cords. Supported voltage ranges from 90V 240V AC (50-60 Hz). Maximum power consumption of the AmpliSpeed (ASC100D- 400D) slide cycler is 100 Watt. 12 of 52 AmpliSpeed system User Guide M10001 Version 4.2

13 04Setting up the AmpliSpeed Optional: Connecting the network The network socket is located at the backside of the display unit (see fig.1). Use standard RJ45 Ethernet cables (not supplied) for connection. Please contact the Beckman Coulter support in case of problems establishing the network connection. Figure 1: Display unit rear view Optional: Connecting the CAN-Bus interface The CAN-Bus connection is useful for easy integration of the AmpliSpeed instrument e.g. in liquid handling platforms. Use a standard sub D cable (not supplied) for connecting the CAN- Bus interface. Please have these details ready when contacting the Beckman Coulter support. ASC100 labels are usually hidden behind other modules (e.g. power supply). Please do not remove modules. 18 05 Getting used to your AmpliSpeed slide cycler 05 Getting used to your AmpliSpeed slide cycler Figure 16: Opening and closing the lid-latch in open position open position Turning the AmpliSpeed on The power switch is located on the right backside of the instrument (see fig. 3). After switching it to on (I) the lid of the power supply blinks a few times and than lights up in blue constantly. Alternating light in red and blue (frequency 1x/sec) during operation indicates a significant error. Please consult the Beckman Coulter support team. Slow blinking light in blue (1x/sec) is a reminder for user interaction or a finished process. Figure 17: Opening and closing the lid-latch in closed position closed position Inserting & removing a glass slide The AmpliSpeed system has been designed for compatibility with AmpliGrid and standard microscope slides. Dimensions of up to 1 mm x 25 mm x 76 mm are possible to fit in the slide bay. To insert a slide please open the plastic lid by moving the grey latch to the right fence (see fig. 16). Then lift the lid to the 90 position. Put the slide on top of the silicon surface and move it to the leftmost edge (see fig. 18). Close the lid and move the grey latch to the leftmost position (see fig. 17). This locks the lid and fixes the AmpliGrid slide to the optimal position directly underneath the drilled holes in the lid for accurate pipetting using liquid handling platforms. Note: If using different slide geometries (e.g. slides with mounted plastic parts) the lid might be left open during operation. In general this is possible without negative effects on the performance of the system. Figure 18: Inserting and positioning a slide holes for automated pipetting Turning the AmpliSpeed off When turning the AmpliSpeed off, it is not necessary to exit the control software first. Please switch the instrument off at the power switch ( O ) located on the right backside of the power supply (see fig. 3) slide fence slide bay latch / slide positioner! Attention! 20 06 Using the Control Software 06 Using the Control Software General information / Login When switching the AmpliSpeed on, the instrument performs a short self-check and starts up with the graphical user interface automatically after finishing the login procedure (see fig. 19). Please login as administrator during the first start of the instrument. Select Administrator by touching it in the selection field and press Login. The instrument comes without a default password for the administrator. For more information about user administration please see chapter 08. Before any kind of creating, editing, loading or running programs you have to select a unit in the left frame of the display (see fig. 20). Figure 19: Login screen of AmpliSpeed slide cycler After the unit selection the AmpliSpeed main window appears (see fig. 21). The slide units attached are represented by 1-4 tabs in the left frame, depending on the instruments configuration. Switching between those units is possible by touching the corresponding tab. The selected unit is highlighted in a darker color and the program loaded to this unit is displayed. A program loaded before switching off the instrument will be available directly after restarting the system. Figure 20: Unit selection after login process In the top frame the pull-down menus for file management and options are available as well as the buttons needed for creating a temperature profile (see appendix A). The main frame shows the loaded program in the idle state, detailed progress information during a PCR run and is the platform for creating new programs. At the left side of the bottom frame direct access to user administration is provided, as on the right side the slide incubation function is located. By touching the numbers or characters on the displays main frame with the input pen (supplied) basic values like time or temperature can easily be changed. Also adding, deleting or copying steps is possible directly in this dialogue. For your convenience running and finishing a program is obtained by touching the start or stop button (see fig. 22) also with your fingers. Please do not use any sharp instruments to touch the display. 21 06 Using the Control Software When you are prompted for changing numbers or characters you can do this by using the up-and-down arrows next to the input field. As a second option a fully functional soft-keyboard pops-up (see fig. 23) on the screen for direct input. By touching the selection fields in the upper row, you can easily switch between capitals, lower case letters, numbers and special characters. If this keyboard interferes with the navigation or the input fields it can be moved on the screen by touching and moving it with the input pen. Please use the scrollbar to move the displayed region on the screen if only parts of the temperature profile are visible. Figure 23: AmpliSpeed soft keyboard Creating a new program Select the active slide unit by touching the corresponding rectangular box in the frame on the left side on the screen. Please ensure that the unit to select is in idle mode. You are not able to create, edit or save programs when an unit is in use (displaying Load, a countdown timer, Heating or Ready ). Please change the unit first by selecting an idle state unit in the left frame or end the running process. Note: The AmpliSpeed internal storage device is limited to 600 temperature steps per program (including repeats during cycles). Please do not exceed this number when creating a program. Otherwise the program will be skipped during the loading process. Figure 24: Opening the File Menu dialogue Select New in the file menu dialogue (see fig. 24). The mandatory End step (see fig. 25) is essential for the AmpliSpeed slide cycler to finish a program run. This step cannot be deleted. When reaching this step all temperature control functions are switched off. The instrument equilibrates to ambient temperature. Adding a new temperature step Press the Temp button at the toolbar. Touch the screen at the End step. In the pop-up window the basic settings will be displayed. Select temperature, time and heating rate (see fig. 26). Note: The AmpliSpeed slide cycler is extremely fast. For some applications it might be useful to reduce heating rates. Please read the manual of e.g. the amplification enzymes for proper settings. Figure 25: Cleared main frame with End step Also a temperature ramp can be defined using the Temp Ramp tab. This feature allows for increasing/decreasing the temperature during the selected time of the step (see fig. 27). When the actual step is not part of a cycle (see section Defining a cycle ) these settings have to be made after inserting the step to a cycle using the Edit button in the toolbar. Clicking OK adds the first step of the profile. Cancel aborts the step. Note: The software will prevent you from setting impossible increments e.g. when reaching temperature limits. In this case, temperature increment values will be set to zero automatically and won t be carried out in the program. For adding an additional temperature step press the Temp button at the toolbar. Touch the screen at any existing step. The new step will be pasted to the left. See section Adding the first temperature step for description of the basic settings. Time increment: Modification of a temperature step for progressive lengthening or shortening of the holding time. Application: Compensation of decreasing enzyme activity during a program. Figure 29: Pause step in temperature profile Temperature increment: Modification of a temperature step for progressive increase or decrease of the temperature each time this step is executed in a cycle. Application: Determining of e.g. annealing temperatures of unknown oligonucleotides Heating rate: Sets a slower-than-maximum rate for the heating process. Cooling ramps are fixed to maximum rate of up to 3 C / sec. The AmpliSpeed slide cycler is extremely fast. 23 06 Using the Control Software Adding additional features Adding other features follows the same routine than described in adding a temperature step. After selecting the button representing the feature to add select an existing step. The new step will be pasted to the left side. If available, popup windows may appear for advanced settings. Pause step Select the Pause button in the toolbar and touch an existing step. The pause step will be inserted to the left (see fig. 29). The AmpliSpeed stays with the settings from the previous step when reaching the pause step. The blue status indicator on the power supply blinks slowly (1x / sec) to remind you for user interaction. Press the Start button to continue. Press the Stop button to abort the program. Figure 30: Selection of signal properties Signal step The AmpliSpeed offers convenient user notification of finished programs or upcoming interactions. Select the Signal button at the toolbar and touch an existing step. In the pop-up window you can select the type of reminder, either an acoustic signal (1-3 rings), an or a SMS (see fig. 30). After clicking OK the signal step will be inserted to the left (see fig. 31). The or SMS function is only available with special configurations and the AmpliSpeed connected to an appropriate network. Please refer to section 07 for setting up these functions or contact the Beckman Coulter support for further information. Figure 31: Signal steps in temperature profile Hold step Select the Hold button in the toolbar and touch an existing step. The hold step will be inserted to the left (see fig. 32). The AmpliSpeed holds the selected temperature for an infinite time. Hold is limited to be the last step of a program for defined storage of the samples until downstream processing. 24 06 Using the Control software Defining a Cycle After the definition of the program steps you can combine a selection or all of those to a cycle. Press the Cycle button at the toolbar. Then touch the screen at the first and the last step of the cycle (see fig. 33). In the following pop-up window you can change these values and define the number of cycles. Steps within a cycle are marked with a black bar. Permitted Values Table 4: Permitted Values Figure 33: Defining a cycle Cycles 1 to 99 in 1 cycle increments Note! Once you have deleted a step or a cycle it cannot be restored. Copy steps or cycles Press the Copy button at the toolbar. Then select the first and the last step to copy. If you want to copy only one step please touch it twice. Also steps from within a cycle can be extracted this way. Paste steps or cycles Press the Paste button at the toolbar. Then select an existing step. The copied step or cycle will be inserted to the left. Delete steps/cycles Press the Delete button at the toolbar and select the step you wish to delete. To delete cycles please touch the black cycle bar. Made a mistake during programming? / Changing existing programs? Cancel selection Press the Cancel button at the toolbar. This deletes a selection in the toolbar e.g. when pressed unintentional. Editing a program step In the temperature profile graph the user can change basic parameters just by touching the time or the temperature of each step or e.g. the cycle number with the input pen. For advanced settings of temperature steps, signal steps or cycles press the Edit button at the toolbar followed by touching the step to edit. In the pop-up window advanced settings will be displayed if available. Please refer to the corresponding section for detailed information. 25 06 Using the Control Software Modifying / saving a program Select the slide unit to use by touching the corresponding rectangular box in the frame on the left side on the screen. If the program to edit has not been already loaded please select Load in the file menu (see fig. 24). Choose the file you want to load and press OK. Change steps or cycles by touching the numbers above and below the temperature graph. For advanced settings select the Edit button in the toolbar. Please refer to Creating a program or the Made a mistake during programming section for information about adding, editing or deleting steps. Save overwrites the existing program after a second user notification. Save as allows storing the program with a different file name or in a different directory on the personal SD card. Figure 34: Saving a program Program names Please select program names according to standard Windows regulations. There is no limitation in characters. Please keep in mind that the graphical user interface only shows up to 15 characters. Longer file names are possible but will be hidden partially on the display. Shared directories Due to regulations in the user administration, standard users have no rights to write, edit and save programs. Those users can be provided with programs by administrators or power users. Please store these files in the <shared data> directory on the SD card. After selecting save in the file menu you are able to switch to the <shared data> directory by selecting this folder in the window and clicking the Ch.Dir button. 26 06 Using the Control software Figure 35: Loading a program Loading / running a program Select the slide unit to use by touching the corresponding rectangular box in the frame on the left side on the screen. For loading a stored program please select Load in the file menu (see fig. 24). Select the program to load and press OK (see fig. 35). After loading the program press the Start button. The program is transferred to the selected slide unit. Please allow a few seconds for the loading procedure. The slide unit will start the program automatically. If a program has not been saved after modification and automated request for saving is displayed. During a program run active steps are highlighted in grey. Start time, stop time, step number and cycle number are displayed in the main frame. A countdown timer shows the time until the end of the run (see fig. 36). Times shown are roughly estimated. After the first cycles the calculation will get more accurate. Please use the scrollbar to move the displayed region on the screen if only parts of the profile are visible. Finishing a program After run time of a temperature profile has finished the status of the slide unity is set to Ready. You will be reminded for your user interaction by the slowly blinking blue light of the status indicator. Please press the Stop button to end the program. Now the slide unit is set back to idle status and is ready for a new run. Temperature range is from ambient temperature to 99 C in 1 C increments. Then press the Incubate button (see fig. 37). Incubation time will be infinite. 28 07 Options Menu 07 Options menu The AmpliSpeed control software offers basic configuration parameters for the hard & software of the instrument. Those functions are only available to administrators. Power users are limited to file space management; standard users are not able to access this menu. Please select the Options button in the toolbar (see fig. 38). Then select the appropriate menu. Figure 38: Options menu Cycler configuration This menu gives a short overview about the status of the slide units attached (see fig. 39). After selecting a unit tab the software and firmware versions and actual status are displayed. This information is useful when contacting the Beckman Coulter support. It is also possible to disable distinct units by deselecting the checkbox. Figure 39: Cycler configuration The calibrate button is only active when having the automated temperature calibration tool connected. Please contact the Beckman Coulter support for further information about temperature calibration. The self-test is automatically carried out during the start-up phase of the instrument and can be repeated by clicking the Self test button. Software configuration This menu displays the AmpliSpeed control software version on the basic screen and allows for advanced settings of PCR (see fig. 40). Figure 40: Software configuration - basic screen PCR settings (see fig. 41) Auto restart within XX min: a user defined auto restart time after power loss can be set. The maximum time is 10 min, as longer periods of pausing without temperature control will interfere with the PCR results. For critical protocols we recommend to decrease this time. Get PCR comment: Activating this check box will result in a comment prompt after starting the PCR. This allows for a short description of the PCR program for GLP integration. GLP conformity: The AmpliSpeed control software offers a full GLP report function in combination with the PCR software. Please refer to section 09 for a detailed description of the GLP feature. 30 07 Options Menu Log Files The AmpliSpeed control software offers convenient user and error logs for GLP / GMP integration (see fig. 46 & 47). These files are automatically saved on the personal SD-card and can either be read out on the touch screen display or transferred to the PC software. For older error logs please use the Archive button, select an archive (error logs are sorted numerically ascending, user logs are sorted by date) and click Show (see fig. 48). Logs can be deleted and saved at a different file path. Changing the view between archive and current is possible by clicking the corresponding button. File Space Management Depending on the usage of the AmpliSpeed instrument, internal RAM might run full after a certain period of time if you do not save the log files periodically on your personal SD card. You will be prompted before entering critical levels of remaining storage. Please use the Management file space dialogue (see fig. 49) for removing older or unused files. The calendar allows to select files to be deleted when older than a date specified. Alternatively log files can be transferred to the personal SD card. Please pick a user from the list by touching the name with the input pen (see fig. 50). If a password has been selected please type it in the password field. Then press the Login button. Changing the user is also possible during running a program. Press the user short name on the bottom left side of the display and select the user you want to login. Programs already running cannot be stopped by other users. Only viewing the status is possible for others. Please re-login to end those programs. Please define a username, short name and password and select the user level. Administrators have full access to all instrument options and programs. Power Users are able to write, edit, save and delete programs but do not have access to deeper instrument options. Users are only allowed to load and run programs from a pre-defined (shared) folder. Administrators or Power user have to save programs there for lower user levels. Press OK to save and Cancel to leave without changes taking effect. Edit (see fig. 53): editing an existing user profile. Please see Add section for further information. Only administrators are able to edit user profiles. Delete (see fig. 54). Deletes an existing user. User once deleted cannot be restored. All programs, log files and settings will be lost. Only administrators are able to delete user profiles. Default admin password : When inserting a new SD-card it is essential to set a default user to get this card functional. Please select an administrator password for this SD-card to get access to administrative functions. 36 10 Maintenance / Warranty / Repair / Disposal 10 Maintenance / Warranty / Repair / Disposal Maintenance The AmpliSpeed has been designed as an almost maintenance free instrument. The surface of the housing can be cleaned from time to time with a soft cloth. Do not use strong detergents or organic solvents. We strongly recommend keeping the surface of the heating plate clean to ensure optimal heat transfer to the slide. If liquids have been spilled, clean immediately with water and/or alcohol using a soft and lint free towel. Do not use abrasive cleaners or hard materials that may scratch the surface of the silicon plate. Clean the air intakes at the bottom of the instrument on a regular base to ensure free airflow using a vacuum cleaner. Blocked air intakes may lead to insufficient cooling performance of overheating of the instrument. Please also read the safety warnings in section 02. Warranty / Repair Beckman Coulter offers a one-year warranty on all AmpliSpeed products. For extended warranty, service contracts or temperature calibration services please contact the Beckman Coulter support or your local representative. The instrument may only be opened and repaired by authorised service personnel. The warranty will be refused in the case of unauthorised servicing or opening of the instrument. In the unlikely case of a malfunction of the AmpliSpeed slide cycler please contact your local representative or the Beckman Coulter support.
