Install National Instrument Measurement and Automation Explorer (MAX) (Available under the master folder or also can be downloaded) 3. Install the camera a. Connect the camera to your computer (AVT Guppy 503B) b. Open MAX (Start all programs National Instruments Measurement and automation) c. Locate camera on MAX (See Figure 1Error! Reference source not found.) and right click on it and select NI-IMAQdx IIDC Digital Camera as the driver Page 1 of 7

4 RUNNING OF THE DATA ACQUISITION AND CONTROL PROGRAM FOR THE MICROSCOPE The data acquisition and control program of the Multispectral Imagine Microscope is written in Labview. The user interface (UI) of the program can be seen in Figure Major step of to be followed 3. Statistics of the preview 4. Preview of the image 2. Recorded setting for different sources 5. Stop execution button Figure 4 - The user interface of the program MAJOR STEPS TO BE FOLLOWED There are 4 straight forward steps to be followed for configuring microscope and acquisitioning images. Once you start the program (by pressing the run button - Figure 5), you need to follow those steps. Figure 5 Run button When program is running, DO NOT stop the program from the menu or using stop button in the key board, instead use stop execution button available in the front panel (see Figure 4). 5 STEP 0: FILE LOCATION AND SAMPLE INFORMATION Once you started running the program, it will prompt you to select a folder where you need to store your images. Usual Microsoft Open / Save As dialog box is the prompt for you and brows for your folder and remember to click SELECT FOLDER button. Then you will be requested to enter name of the sample (Figure 6). If you need to configure acquisition settings according to the bright and dark reference, remember to click the button This is a reference (button caption should be yes ). Figure 6 - Sample name request dialog box STEP 1: HARDWARE SETTING Under this step you are expected to select appropriate device and channels for the DAQ card and camera. If you select Auto mode (see Figure 7), the program will select the device automatically, otherwise you need to select appropriate configuration BEFORE running the program. Detection of camera is not automated. If you get any error message from the program you need to change the camera settings BEFORE you run the program again. Most of the time you will not need to do anything with this tab. If there is no any problem program will automatically displays tab 2. Figure 7 - Hardware settings tab Page 5 of 7

6 STEP 2: ACQUISITION SETTINGS Mean and the variation of the pixel values Warning indicator and warning threshold Histogram of the pixel values Camera settings for the current light source Settings loading to the database Selection of the wavelengths of LEDs Start acquisition for selected settings Figure 8 Acquisition settings tab STEP 3: ACQUIRING IMAGES When you press the Start button in step 2, the program will starts capturing images. If you have selected reference as your first measurement, program will ask you to change the target for your references for all geometries. Then it will ask for the sample to place on the sample table. You can take as many as sample images. Once you finish with taking images you can press cancel, when program ask for next sample. While images are being acquired, you can see the histogram and preview of that image. Page 6 of 7

7 Figure 9 Acquisition information tab FREQUENTLY ASK QUESTIONS (FAQ) 1. WHERE MY IMAGES ARE RECORDED AND WHAT IS THE FOLDER STRUCTURE? The images are saved in a folder named as your sample name, in the folder that you have selected when program is started. Inside that folder, there will be one folder called reference and several folders starts with sample0. Inside the reference folder there are six sub folders TB, RB, SB, TD, RD and SD (T Transmission, R Reflection, S Scattering, B Bright reference, D Dark reference) and in sample folder there are three folders TS, RS and SS. 2. WHY CAN T I VIEW IMAGES USING MICROSOFT FAX VIEWER The images you saved are 16-bit images, which are not compatible with Microsoft Fax Viewer. You can view then on matlab or you can use ifanview software. Page 7 of 7
