1 Agilent Installation Infinity Checklist II Preparative HPLC System Thank you for purchasing an Agilent instrument solution. This checklist is used by the installing engineer to ensure that the instrument and associated systems are correctly installed, upgraded and functioning as designed in your facility. This checklist will be completed at the end of the service and provided to you as a record of the installation. Customer Information Customers should provide all necessary operating supplies upon request of the engineer. Service Engineer s Responsibilities Only complete/printout pages that relate to the system being installed. Complete empty fields with the relevant information. Complete the relevant checkboxes in the checklist using a X or tick mark in the checkbox. Complete Not Applicable check boxes to indicate services not delivered, as needed. Complete the Service Review section together with the customer. Additional Instruction Notes A checkout test is available. The delay calibrant ( ) can also be used as checkout solution when injected onto a column and run with a gradient. The result should show three colored compounds which are collected in three different fractions. At the end of the installation, run the System Test (described below). System Test 1 Ensure that a ZORBAX SBC18 PrepHT 21.2 mm (or 9.4 mm) 50 mm, 5 µm column is connected, and that the LC system is properly primed. 2 Check that the mobile phase and needle wash solvents previously used by the customer are compatible with the solvents used for the System Test. If not, use an appropriate intermediate solvent to prepare the system.

Mode: Threshold Peak 3, Sudan Orange (M=214.22) MNa 203 neg (double charge) MNa 543 neg, MNa+H 545 pos, M+H 567 pos M+H 215 pos 1.5 ml/min with only 33 % of this entering the MSD (Teeline split) 4 Ensure that 3 peaks are observed and three fractions are collected (yellow, blue and orange) and enter the results in the results table at the end of this checklist.

4 General Preparation Unpack/verify the condition and completeness of shipment. For discrepancies, use the table below. 12 G6100 Series Single Quadrupole Mass Spectral Detector Using two people, place the MSD on a stable bench or table. Make sure adequate space is available to operate and maintain the instrument. Install the spray chamber drain bottle. Install the rough pump and fill with oil (if applicable). Install the Agilent TS800 Dry pump (if applicable). Install the TS800 Scroll pump ship kit (if applicable). Connect the MSD vacuum hose to the rough pump. Install the rough pump exhaust KF fitting adapter. Connect the other end of the exhaust tubing hose to the rough pump exhaust vent. Connect the spray chamber exhaust tube between the desolvation assembly and 1" drain bottle fitting. Connect a length of 3/4" exhaust tubing to the drain bottle vent fitting. Connect the other end of the exhaust tubing hose to a separate exhaust vent from the rough pump. Attach the waste line from the MSD to the drain bottle 1/4" fitting. Connect the LC leak drain system by moving the MSD right front cover. Install RMSN4 nitrogen gas filter(s) using 1/4" PTFE tubing. Install RMSN4 filter between nitrogen supply for the MSD. Secure gas filter(s) vertically to suitable support(s). Turn on the nitrogen supply, set the regulator between psi ( kpa), and check for leaks. Connect the rough pump power cord to the MSD. Connect the MSD power cord to the power outlet. Connect the LAN cable to the MSD LAN port. Install the Remote cable between one LC module and the remote port of the MSD. Install the APCI enablement kit (only applicable for APCI or Multimode source). Install the Multimode enablement kit (only applicable for Multimode source). Uninstall the spray shield and remove the red GC septum from the capillary entrance. Reinstall the spray shield. Install the ES source (if applicable). Attach the source to desolvation assembly. Install and align the standard Spray Shield (G ) on the MSD. Connect the 1/8 nebulizer gas line from the MSD to the nebulizer gas fitting. Connect LC tubing from Flow Modulator to nebulizer ZeroDeadVolume fitting. G6125B/C and G6135B/C: Add G ESI tuning calibrant.

13 Install the APCI source (if applicable). Attach the source to desolvation assembly. Install and align the standard Spray Shield (G ) on the MSD. Connect the 1/8 nebulizer gas line from the MSD to the nebulizer gas fitting. Connect LC tubing from Flow Modulator to nebulizer ZeroDeadVolume fitting. Connect the APPI heater cable to the MSD Vaporizer Heater connection. Connect the male APPI power and communication cable to the DB9 connector on the source. Connect the female APPI power and communication cable to Serial B port on the MSD SmartCard. Add G APCI/APPI tuning calibrant. Install the Multimode source (if applicable). Attach the source to desolvation assembly. Install and align the Multimode Spray Shield (G ) on the MSD. Connect the 1/8 nebulizer gas line from the MSD to the nebulizer gas fitting. Connect LC tubing from Flow Modulator to nebulizer ZeroDeadVolume fitting. Connect the APCI high voltage cable to the MSD at the Auxiliary Module Corona connection. Connect the Multimode high voltage cable to the MSD at the Auxiliary Module Multimode connection. Connect the Multimode heater cable to the MSD Vaporizer Heater connection. G6125B/C and G6135B/C: Add G APCI/APPI tuning calibrant. Install the Jet Stream source (AJS) (if applicable). Attach the source to desolvation assembly. Install and align the AJS Spray Shield (G ) on the MSD. Connect the 1/8 nebulizer gas line from the MSD to the nebulizer gas fitting. Connect LC tubing from Flow Modulator to nebulizer ZeroDeadVolume fitting. Connect the 1/8 AJS sheath gas line from the Agilent Jet Stream to the MSD sheath gas fitting. Connect the AJS nozzle voltage cable to the MSD at the Auxiliary Module Multimode connection. Connect the AJS Sheath Gas heater cable to the MSD Vaporizer Heater connection. G6135B/C: Add G ESIL tuning calibrant. Turn on the MSD and allow the system to pump down. Verify the operation of the MSD Perform a Dual Polarity Autotune on the G6125B or G6135B (if applicable). Perform a Dual Polarity Checktune on the G6125C or G6135C (if applicable). Perform a background scan check from m/z 100 to 1000 at 0.4 ml/min: G6125B/C: TIC baseline should be less than counts. G6135B/C: TIC baseline should be less than counts.

14 G6125B/C: Perform a ES positive ion SIM checkout and confirm RMS signaltonoise of 30:1 G6125B/C: Perform an APCI positive ion SIM checkout and confirm RMS signaltonoise of 15:1 G6135B/C: Perform a ES positive ion SIM checkout and confirm RMS signaltonoise of 100:1 G6135B/C: Perform a ES positive ion Scan checkout and confirm RMS signaltonoise of 100:1 G6135B/C: Perform an APCI positive ion SIM checkout and confirm RMS signaltonoise of 50:1 G6135B/C: Perform an APCI positive ion Scan checkout and confirm RMS signaltonoise of 50:1 G6135B/C: Perform an AJS positive ion SIM checkout and confirm RMS signaltonoise of 500:1 G6135B/C: Perform an AJS positive ion Scan checkout and confirm RMS signaltonoise of 500:1 Fill out instrument logbook. Flush the LC overnight with 0.5 ml/min of 70:30 ACN:H 2 O with 0.1 % Formic Acid using the following spray chamber parameters: ESI source: Nebulizer Pressure 40 psi, Drying gas flow 10 L/min, Drying gas temperature 350 C. APCI or APPI source: Nebulizer Pressure 40 psi, Drying gas flow 5 L/min, Drying gas temperature 350 C, Vaporizer temperature 450 C. Multimode Source: Nebulizer Pressure 40 psi, Drying gas flow 5 L/min, Drying gas temperature 350 C, Vaporizer temperature 200 C. AJS Source: Nebulizer Pressure 40 psi, Drying gas flow 5 L/min, Drying gas temperature 350 C, Sheath gas temperature 350 C, Sheath gas flow 10 L/min, Nozzle voltage 0 V. Make sure the Switch Stream option is set to MSD. Bake out the system for at least 2 hours; preferably, bake out overnight. After the overnight flushing, flush with 100 % isopropanol for 30 minutes at 1 ml/min. Flush with 50:50 Methanol:Water for 30 minutes at 1 ml/min. Install the 2.1 x 50 mm x 1.8 µm SB18 Rapid Resolution column. Flush with 100 % methanol for 1 hour at 1 ml/min. Prepare LC mobile phase for positive ion verification; 70:30 ACN:H 2 O with 0.1 % Formic Acid Connect 1 liter of checkout solvent to Channel A and flush for 1 hour at 0.5 ml/min. Record system serial numbers and fill out installation documents while flushing.

15 Preparing Performance Evaluation Samples Use 5 ng/µl positive ion (reserpine) standard G2423A (P/N G ) and dilute to appropriate concentration. G6125B/C: SIM: Prepare a 1:5000 dilution (1 pg/µl) G6135B/C: SIM: Prepare a 1:5000 dilution (1 pg/µl), Scan: Prepare a 1:500 dilution (10 pg/µl) Perform a standard instrument verification for the installed source. If more than one source is present, perform verification with each source (see source specific section(s) for details).
