Instructions for Use MTP TLC Adapter Accessory to enable the measurement of thin layer chromatography (TLC) plates and large-format glass slides in matrix-assisted laser-desorption/ionization (MALDI) mass spectrometers with ScoutMTP source 1) The product is for research use only. It is not for use in diagnostic procedures. Product description The MTP TLC Adapter (see Figure 1) is a dual-use adapter for TLC MALDI 1) and for MALDI Imaging applications to be measured on a Bruker ScoutMTP source MALDI instrument. 75 x 50 mm format TLC plates can be mounted on this adapter as well as the BigSlides for MALDI Imaging (# ) with the same format. 2 For TLC MALDI applications we recommend the use of 75 x 50 mm aluminium backed TLC silica gel plates (see recommendations in Ordering Information). Please note: Do not use TLC plates with glass (or other non-conductive) backing. MALDI TOF requires electrically conductive sample plates. n-conductive plates will cause electrostatic charging problems. For MALDI Imaging applications use dedicated 75x50 mm glass slides (BigSlides for MALDI Imaging, # ). These slides are equipped with a transparent electrically conductive ITO coating on one side. The coated side of the slide is easily visible when the box is open and can be checked with an ohmmeter. Make sure that you put the tissue sections on the coated side of the slides! Please note: Do not use uncoated glass slides. They will cause electrostatic charging problems. Ordering Information Product Unit Part. Some older versions only recognize the MTP TLC Adapter after installation of a free (see Table 1). Compass 1.1 and older do not support the TLC target. 3 Mounting a TLC plate Place the adapter face-down on a table (see Figure 2a). First insert the TLC plate, then insert the metal stabilization plate and lock the retaining tabs following the procedure shown on Figure 2b to Figure 2f. a b c d e f Figure 2 Mounting of a TLC plate on the MTP TLC Adapter Mounting glass slides for MALDI Imaging The MTP TLC Adapter is also suitable for glass slides of a 75 x 50 mm format and suitable thickness for MALDI Imaging applications (e.g. BigSlides for MALDI Imaging, # ). Place the glass slide in the adapter as described for TLC plates in the above procedure and also mount the stabilization plate. The electrically coated side (with the tissue section applied) must face the front side of the adapter target! There is no risk that the tissue on the glass slide will touch the table as the protruding screws on the front side of the adapter act as a spacer. Matrix Application We recommend the use of the Bruker ImagePrep spray preparation device (# ) for matrix coating. Dedicated methods for TLC sample preparation are available for download from our homepage. Contact Bruker Support to obtain the relevant. MALDI Measurements TLC plates have a porous surface. This can lead to significantly prolonged pumping time when introducing TLC plates into the mass spectrometer. As a result, a timeout error may occur. If this happens, please repeat the introduction procedure. If this becomes a continuous error, please contact maldi.sw.support@bruker.com for assistance. Page 3 of 5

4 Teaching and Spot Localization For TLC MALDI applications use the metric scale on the adapter target to read out the lane positions. The coordinates can be entered directly into the TLC MALDI tool without any need for further teaching (see Figure 3). For MALDI Imaging applications use Bruker fleximaging to teach the tissue sample position. This provides a convenient 3-point teaching procedure which matches the microscopic image of the tissue section to the actual target position in the instrument (for details please refer to the online help for fleximaging). Coarse navigation on glass slides or TLC plates is simplified by using the 384 array coordinates: 1) put an MTP target cover plate (which is supplied with the adapter) onto the slide adapter, 2) label the spots of interest or the outer contour of the sections on the cover plate, 3) put the cover plate onto a conventional 384 target and read out the coordinates.
