3 1. Principle, Application and Features 1.1 Principle Substances in solutions under radiation of light absorb light, and this kind of absorption is selective. Different substances have their own absorption spectrums. Therefore, when homogeneous light passes through a solution, its energy will be reduced due to absorption. The degree of energy reduction is in proportion to the concentration. UV754N UV-visible spectrophotometer is a single-beam spectrophotometer with simple construction and ease of use, for qualitative and quantitative analysis of samples based on the aforementioned principle. As to the principle of measurement and analysis, the parameters meet the Lambert-Beer law with a specific range of concentration. Installation 3.1 Installation Conditions Place the instrument on a firm and leveled bench meeting specific lab environment requirements and the following conditions: Lab room temperatures between 5 C 35 C, and relative humidity of not higher than 85%, Power supply of AC 220 V 22 V, 50 Hz 1 Hz, with good earthing, Free of direct sunlight, vibration, strong air flows and corrosion by corrosive substances, and Away from strong magnetic fields, electric fields, and electrical devices generating high frequency waves. Note: Use AC stabilized voltage power supply of 500 W or above in the case of noticeable voltage fluctuation. 3.2 Inspection upon Unpacking Inspection Please check the integrity of the external package before unpacking. Contact the transport firm if there is any damage or incompleteness in the package Checking Open the packing along the sealing seams (Keep the external package for possible use in the future). Check the instrument and spare parts against the list of spare parts. Contract the local dealer or directly the manufacturer if there is any inconsistency. Page 5

8 4.2 Keypad 1. T / A / C/F: Press this key to switch between the display modes of T (transmittance), A (absorbance), C (concentration) and F (concentration factor) for the data display, and the mode is indicated in the LCD. 2. Enter/Print: This key has 2 functions: a) Start printing and send data to a computer via the RS232C serial port (one-way data transmission). b) When the instrument is in the F status, the key is for confirmation of the concentration factor which has been entered or modified, i.e. the current F value, and automatically calculates the current F value (F=C*A). c) When the instrument is in the C status, the key is for confirmation of the standard sample concentration which has been entered or modified, i.e. the current C value, and automatically calculates the current F value (F=C/A). 3. /0%: This key has three functions: a) Zeroing: This function is only valid in the T status. Open the sample chamber cover and press the key. The indication should be b) Decrement: This function is valid in the F status. Press this key to automatically decease the F value by 0.1 for each pressing. Press and hold this key to decrease the value by 1 at a certain interval. If the F value reaches 0000, the value will become 1999 when this key is pressed again to restart decreasing. b) Decrement: This function is valid in the C status. Press this key to automatically decease the C value by 0.1 for each pressing. Press and hold this key to decrease the value by 1 at a certain interval. If the C value reaches 0000, the decrement is stopped. 5. /100%/0A: This key has three functions: a) Adjustment full range / absorbance zeroing: This function is only valid in the T and A status. Close the sample chamber cover and press the key. The indication should be and b) Increment: This function is valid in the F status. Press this key to automatically increase the F value by 0.1 for each pressing. Press and hold this key to increase the value by 1 at a certain interval. If the F value reaches 1999, Page 7

9 the value will become 0000 when this key is pressed again to restart increasing. b) Increment: This function is valid in the C status. Press this key to automatically decease the C value by 0.1 for each pressing. Press and hold this key to increase the value by 1 at a certain interval. If the C value reaches 1999*A, the increment is stopped. 4.3 Display Test mode indication: T (transmittance), A (absorbance), C (concentration), and F (slope). 2. Data display Page 8

10 5. Operation 5.1 Basic Operation Warming upon Power-on After the instrument is connected to power supply, the microcomputer conducts self-check. After the LCD shows 752, the instrument enters working status. Blow, the default mode shown on the screen is T. Note: The warning upon power-on should last for no less than 30 minutes for the inside of the instrument to reaching thermal equilibrium Change Wavelength Use the wavelength knob to change the wavelength setting of the instrument, and select the desired wavelength with the scale of the wavelength window. ATTENTION IF THE WAVELENGTH RANGE IS 330 NM 370 NM, PLEASE PUT THE CYLINDRICAL FILTER ONTO THE LENS CYLINDER OF THE SAMPLE CHAMBER. THE FILTER IS NOT REQUIRED FOR OTHER WAVELENGTHS Place Reference and Test Samples Select a colorimetric plate. Place a reference/test sample onto the sample holder. Use the sample holder level to change the position of the sample. When the level is in place, you hand will feel the sense. Gently pull and push the level to ensure the position is right in place Adjust 0%T and 100%T/0A To ensure the instrument to enter correct testing state, press the /0% and /100%/0A keys to conduct zeroing and full range and absorbance zero adjustment, after the test wavelength has been changed or the instrument has been in use for a period of time. 11 5.1.5 Select Display Mode 752N UV-Visible Spectrophotometer Operating Instructions The instrument has four display modes. The initial mode after power-on is the transmittance mode (T). 1. Transmittance (Trans) 2. Absorbance 3. Concentration (Conc.) 4. Concentration factor (FacTor) Direct Concentration Reading and Concentration Factor 1.Direct concentration reading setting Press the T / A / C/F key to select direct concentration reading (c). Press /0% and /100%/0A to enter the desired standard sample concentration. Put in the standard sample and press Enter/Print or Enter/Print, and then test the unknown sample. The concentration factor will be automatically changed accordingly after the calculation. You may view the F value and record it. The concentration factor can be directly used in subsequent tests for the same samples, without calibration with the standard sample. Note: Put in a standard sample before direct concentration reading setting. 2.Concentration factor setting Press the T / A / C/F key to select direct concentration reading (F). Press /0% and /100%/0A to enter the desired concentration factor. Press Enter/Print or Enter/Print, and the display automatically returns to the direct concentration reading (C) mode, and then test the unknown sample Print When the instrument in the T, A or C display mode, connect it with a serial printer via the RS232C port. Press Enter/Print to print the displayed data. Note: You need to purchase a separate printer Exchange Data with RS232C Serial Port The instrument is able to communicate with a PC installed with the UVWin7 software developed by the manufacturer via the RS232C serial port. Operations 6.1 Test Transmittance / Absorbance 1 Warming 2 Select test wavelength 3 Place reference and test samples 4 Select T/A mode 5 Adjust 0%T and 100%T/0A with the reference sample 6 Test the test sample 7 Record or print data 6.2 Application of Direct Concentration Reading If the sample to be analyzed is stable and the standard curve nearly passes the origin, it is unnecessary to use the relatively complex standard curve method to determine the concentration of the sample to the tested. You may use the direct concentration reading method for quantitative testing. 1 Warming 2 Select test wavelength 3 Place reference and standard samples 4 Adjust 0%T and 100%T/0A with the reference sample 5 Select C mode 6 Enter the concentration of the standard sample. 14 7. Maintenance and Failure Identification 7.1 Daily Maintenance 1. To ensure the stability of the instrument, it is recommended to Use AC stabilized voltage power supply of 500 W or above in the case of noticeable voltage fluctuation. 2. When the instrument is stopped, switch off the power button on the instrument, and then disconnect it from the mains. 3. Use a soft cloth wetted with warm water to clean the instrument case. Do not use ethanol, ether, acetone or other organic solutions. Place the dust cover on the instrument after use. 4. Clean the sample chamber and colorimetric plate holder after each time of use, to prevent corrosion of instrument parts caused by the samples. 5. Use petroleum ether to clean colorimetric plates after each time of use, use lens tissue to wipe the colorimetric plates clean, and place them in colorimetric plate boxes for future use. 6. Periodically check the wavelength of the instrument according to metrology rules, to ensure the availability and measurement accuracy. 7.2 Light Source Change The 754N UV-visible spectrophotometer uses a Philips 12v/20w inserted tungsten halogen lamp and DD2.5 deuterium lamp. Observe the following procedure for replacement: Tungsten halogen lamp 1. Turn off the instrument and disconnect the power cord from power supply; 2. Rotate off the fixing screws on both sides of the case and the wavelength knob; open the case rightward; 3. Remove the protective guard plate from the lamp chamber; 4. Wear clean gloves to pull off the old lamp, and insert the new lamp; 5. Switch on the instrument; select wavelength of 500 nm to check if the filament image is focused on the Incident slit. If no, loosen the fixing screws of the light source component; slightly adjust the light source component to make the filament image be focused on the Incident slit; at the same time, check the data window s data to ensure the displayed data the maximum values, before tightly fixing the screws. 6. Close the instrument case, and rotate tight the wavelength knob. Page 13

15 Deuterium lamp 1. Turn off the instrument and disconnect the power cord from power supply; 2. Rotate off the fixing screws on both sides of the case and the wavelength knob; open the case rightward. 3. Remove the protective guard plate from the lamp chamber; 4. Wear clean gloves to pull off the old lamp, and insert the new lamp (Note the wire colors of the lamp); 5. Switch on the instrument. Select wavelength of 240 nm to check if the light spot is focused on the Incident slit. If no, loosen the fixing screws of the light source component; slightly adjust the light source component to make the light spot be focused on the Incident slit; at the same time, check the data window s data to ensure the displayed data the maximum values, before tightly fixing the screws. 6. Close the instrument case, and rotate tight the wavelength knob. Note: Replace the tungsten halogen lamp only after the instrument is disconnected from power supply and the tungsten halogen lamp has been cooled down. Pay attention to wire colors when replacing the deuterium lamp. Do not observe the lamp with naked eyes for long. 7.3 Troubleshooting Trouble Cause Solution 1. The instrument does 1. Calibrate the wavelength 7.4 Wavelength Correction The 752N UV-visible spectrophotometer uses the characteristic absorption peaks (to be set and calibrated by the user) of didymium filter at 529 and 808nm to calibrate and correct wavelengths in a point-by-point manner. The optical splitting system uses a grating as the dispersion component. Because its dispersion is linear, the wavelength gradations are linear too. If the scale wavelengths obtained with the above point-by-point method and the characteristic absorption wavelengths of the didymium filter are larger than the specified error range (within 2nm), remove the wavelength knob, open the case, rotate the three positioning screws on the wavelength scale, to make the indication at the characteristic absorption wavelength, and then rotate tight the screws. Storage and Free-Service Period The manufacturer undertakes to repair the instrument for free if it fails to work due to defective workmanship within 12 months following the purchase date, provided that the damage is not caused by intentional act.
