3 GENERAL The Quintessential Injector (Catalog No ), manufactured by Stoelting, is designed for the infusion and/or withdrawal of pico/nano/microliter liter volumes via a Hamilton syringe ( microliters). The following items included are: Controller Unit Syringe Drive Motor Universal Power Supply Operation Manual If the instrument is damaged or, after having tested it, fails to meet rated performances, notify the carrier and our company immediately. 3

4 Before Applying Power Take a look at the back of the Electronic Unit, on the left of the Electronic Unit backpanel, which encompasses from left to right the main power switch, the inlet AC connection of the power source (120V or 220V), Syringe Drive Motor Connection, and Serial Cable Connection for Computer Interface. Connect the DC output cord from the Universal Power Supply to the AC Power Connection as illustrated in Figure 1 and plug into an electrical outlet. You will also need to connect the Syringe Drive Motor cable. If this cable is not connected the unit will not start. Rather, an error message will be displayed stating Motor not connected and to turn the power off connect the cable and try again. V Clamp Adaptor for mounting to Stereotaxic apparatus Syringe Plunger Mount and Drive Block Connecting Syringe Motor Drive Begin by connecting the cable between the Syringe Drive and control unit. If this cable is not connected properly, the unit will not start. Rather, an error message will be displayed stating Motor not connected turn the power off connect the cable and try again. 5 Loading the Syringe The syringe drive has a universal adaptor for most syringe types (including all Hamilton 700, 7000, and Gas-Tight Series syringes, ranging from 0.5 to 250 µl sizes) After loosening the thumb and hex screws insert the syringe barrel into the syringe mount. Using the Fast Forward/Reverse (<< >>) icon (see Fast Forward/Reverse section below for more details) position Syringe plunger mount and drive block so that the syringe plunger knob can be inserted. Once the plunger and syringe barrel are in place retighten the hex screw and make appropriate adjustments by tightening the thumbscrew (see Figure 2 above). Attaching the Pump The Quintessential Injector is not designed as a stand alone item. Rather the unit is designed to be paired with either a Stereotaxic apparatus or micromanipulator. As such the Syringe Drive Motor is equipped with a V clamp adaptor for mounting to a Stereotaxic apparatus (in place of the standard probe holder) and a Connection/Positioning Bar for micromanipulator mounting (see Figure 2). Please note that the position and orientation of the V clamp on the Connection/Positioning Bar is fully adjustable to accommodate a wide rage of pump placements and directions. OPERATING INSTRUCTIONS When the power is turned on, the startup screen to the left appears. This screen will remain on for approximately five second and contains details /specifications relative to the QSI Additionally, this screen calls attention to the What do I do? feature in the bottom right hand corner of the display. This button can be pressed whenever assistance is needed. Main Screen The Startup Screen will be followed by the Main Screen (pictured to the right) From this screen you will input and/or change the values necessary for your specific application. Using your index finger or the stylus supplied with the machine you can modify Syringe I.D., Volume, Rate, and Mode (specific instructions for each of the above follow). 5

6 Additionally, on the main screen you will find Start Pressing this icon will Start the pump (see START below for more details). Fast Forward/Reverse Pressing the << icon will fast reverse the pump (plunger drive block) while the >> icon will fast forward the pump (plunger drive block) Tools Pressing the tools icon will open the screen to the left Using this screen you can change Contrast and Brightness (by adjusting the slide rule), Toggle Polarity (choose light field versus dark field), Calibrate the touch screen and select Power up options. In the Power up options you can select to go directly to the pumping screen following power failure. Turning this feature On will continue a no target volume session following such an event What do I do? Pressing the What do I do? Icon on any screen will give you helpful prompts for whatever screen you are currently using. An example of this is demonstrated in the screen capture to the left Syringe I.D. (inner diameter) In order to calibrate the control unit the inner diameter (in mm) of the syringe to be used must be entered. This can be accomplished by pressing the Syringe I.D. icon. Once the icon is selected the following box will appear to aid you in imputing this diameter. There are 2 methods for inner diameter entry. 6

7 1. Direct Entry Using the up/down arrows ( ) will change the inner diameter entry sequentially starting in the 3 rd decimal position. To speed up this process you can use the left/right arrows ( ) to position the specific digit to be changed xxx.xxx (note that the capitalized X indicates the numerical positioned that will change). Now using the up/down arrows ( ) will change values in that position. You can also directly dial the I.D. value using the Calculator icon Once the icon is selected a screen typical of a calculator will appear. Use this screen to directly input the appropriate value. 2. Syringe Library Selecting the Library icon (shown in the bottom left of the box above) will allow you to choose from a range of Hamilton syringes ( ul). Using the left/right (<>) arrows simple identify the syringe model and volume of the appropriate syringe. Once the correct syringe is identified press OK. The inner diameter for that particular syringe will then be displayed in the Syringe I.D. position Volume In order to ensure that the proper volume is distributed it is necessary to select, from the main screen, whether you will infuse/withdraw ul or nl volumes. Once this is determined you are prepared to enter the appropriate volume. As with inner diameter the syringe volume can be changed using the up/down arrows ( ). Pressing these arrows will change the volume to be delivered sequentially starting in the 3 rd decimal position. Or the process can be sped up using the left/right arrows ( ) to position the specific digit to be changed xxx.xxx (note that the capitalized X indicates the numerical positioned that will change). Using the up/down arrows ( ) will change values in the specified position. 7

8 As with I.D. you can also directly dial the Volume value using the Calculator icon. Once the icon is selected a screen typical of a calculator will appear. Use this screen to directly input the appropriate value. Additionally, you can select the No Limits faucet icon if no target volume is required Rate per Minute Pressing the Rate per min button from the main screen will bring up a screen similar to what is shown when either the Syringe I.D. or Volume buttons are pressed. Using the up/down arrows ( ) will change the volume to be delivered sequentially starting in the 3 rd decimal position. Alternatively, using the left/right arrows ( ) you can position the specific digit to be changed xxx.xxx (the capitalized X indicates the numerical positioned that will change). Using the up/down arrows ( ) will change values in the specified position. A third option is to directly dial the Rate per minute value using the Calculator icon. Once the icon is selected a screen typical of a calculator will appear. Use this screen to directly input the appropriate value. Mode The Quintessential pump can be use to either Infuse or withdraw set volumes. Pressing the mode button will toggle between these two options allowing you to select the operation you require for your particular application 8

9 START Once you have entered appropriate values in the required fields (Syringe I.D., Volume, Rate per min, and Mode) for your particular application pressing the Start button from the main screen will initiate the program and take you immediately to the following progress monitoring page. The Monitoring Page allows you to watch a representative display of your infusion or withdrawal in progress. Flow direction Depending on the mode selected either the (withdraw) or (infuse) arrow will be flash Progress Indication A graphic representation of a syringe allows you to monitor the progress of your application. Additionally, the percentage of the total volume (that has been dispensed or withdrawn) is shown. Status of the pump is identified on this page as either Pumping, Stopped or Done Stop from the monitoring page the option to Stop the pump at any time can be easily accomplished. Pushing the Stop icon will halt the progress of the pump, freezing the progress of the application. Once the application is stopped the options to Resume (start the application from its stalled point) or New (return to the previous page where the application can be changed or started from the beginning) are displayed. CLEANING The controller unit and touch screen can be cleaned by using a mild detergent and water; wiping with a cloth or paper towel. This will not damage the touch screen. 11 QSI COMMANDER SOFTWARE The QSI Commander Software is designed to add flexibility and versatility to the Stoelting Quintessential Injector. This comprehensive software allows the syringe pump library to be programmed, modified and expanded. Additionally, the software can be used to visually monitor (via a webcam) and remotely control the Quintessential Injector. The Commander also features a Library Log where information relative to the Quintessential s use (such as treatment, flow rate, volume, reaction, etc.) can be entered and maintained for future reference. The QSI commander contains its own help Menu and How To instructions simply accessed by clicking on HELP or the question mark (?) icon. To get started you need only insert the accompanying CD into your PC s CD drive and follow the installation instructions. 11
