1 MyProject Web Portal User Guidelines Placing an Order with Excel - Tubes Log in to the MyProject Web Portal Log in to the MyProject web portal at: To view in French or German, click on Language in the top right hand corner and then choose the appropriate language. If you are a new user, create your account by clicking on Setup and completing all of the required fields. Once complete, you will receive an with your username and password to confirm your registration. Ordering CapMarkers CapMarkers are bar code stickers, provided free of charge by GENEWIZ, and are required if you are sending in your own primers. Order CapMarkers by clicking on New in the My Capmarkers orders field of the dashboard. Once you order CapMarkers online, they will be sent to you by post. Once received, click Acknowledge receipt from the dashboard - your CapMarkers are now ready to use to identify your tubes of primers. Downloading The Excel Template File To download the Excel file to place your plate order go to the Download menu at the top of the screen Select the correct Excel file for tubes Tube sequencing Excel template
2 Warning: To Enable Content Click the red protected view Click Edit Anyway Click Enable Content in the security warning
3 Filling in the Excel file (Columns A - K) Notes There are three tabs My sequences, Details and Log. All must be present when you upload the excel sheet. Do not modify the details sheet. Each row is for one sequencing reaction including one DNA sample and one primer Column A - Reference Select the service required, either Express Single or Express Consensus If there is a well you require to be left empty, please leave the entire line blank. Do not fill any cells. Column B - Sample Name Enter the name of the sample submitted 15 characters max including spaces and symbols (-, / ) If an old sample stored by GENEWIZ is to be reused, fill in the original sample name and place in brackets the GENEWIZ Online reference number (COLXX-XXXX) of the first order e.g.: TEST1(COL ) Do not fill any other information in relation to the sample (nature of the sample, capmarker, concentration, size, volume, option) Column C - Nature of the Sample Select PCR, Plasmid or BAC Warning: To Synchronize Capmarkers Navigate to Add Ins in your Excel file Click Synchronize Capmarkers Enter your username and click OK To finalize synchronization click OK Column D - Capmarker Sample Select the capmarker reference going to be used for each sample. Column E - Vector Mandatory if plasmid Select the required vector, if the required vector is not available select other and use the comment box before submitting the order online to enter the vector name Column F - Sample Concentration Enter the most accurate concentration for each of the samples Column G - Size of the Construction Enter the most accurate size of the construction for each of the samples Column H - Volume Enter the most accurate volume for each of the samples Column I - Option PRP Optional for plasmid sample: Chargeable if required enter: PRP Column J - Option PUR Optional for plasmid sample: Chargeable if required enter: PUR Column K - Option PRP Mandatory if the PRP option is required Select the antibiotic name for your vector Click on the hyperlink to view antibiotics for common vectors If the antibiotic is not referenced please put other and use the comment box before submitting the order online to enter the antibiotic name
4 Filling in the Excel file (Columns L - T) Column L - Antibiotic Concentration Mandatory if the PRP option is required Enter if there is specific concentration your antibiotic should be used outside of normal guidelines Column M - Primer Name Select the required Universal Primer from the dropdown menu. Do not enter any other primer fields: Capmarker Primer, Degenerated Primer, Primer Sequence, Primer TM, Primer Volume, Primer Synthesis. Proceed straight to Column V If you are sending Template and Primer mixed together please enter PREMIX. Do not enter any other primer fields: Capmarker Primer, Degenerated Primer, Primer Sequence, Primer TM and Primer Volume. Proceed straight to Column V If you are sending your own primer enter the primer name If the primer is to be synthesised by GENEWIZ enter the primer name Column N - Capmarker Primer Select the Capmarker reference going to be used for each primer If an old primer stored by GENEWIZ is to be reused, fill in Column O with the primer name and place in brackets the COL reference number (COLXX-XXXX) of the first order, e.g. PRIMER1(COL ). Do not enter any other primer fields: Degenerated Primer, Primer Sequence, Primer TM, Primer Volume, Primer Synthesis. Proceed straight to Column T Column O - Degenerated Primer If primer contains degenerated bases, please fill the column with an X Column P - Primer Sequence Mandatory if primer synthesis is required Enter the sequence of primer to be synthesised Column Q - Primer Tm (ºC) If you are sending your own primer enter the Tm of your primer If the primer is to be synthesised do not fill in Column R - Primer Volume If you are sending your own primer enter the volume of primer you are sending If the primer is to be synthesised do not fill in Column S - Primer Synthesis Chargeable Mandatory if the primer requires synthesis, enter an X Column T - Service Options and Free Service Options Enter the appropriate letter (BDHSVTIM) if any of the below services are required Service Options: Free service options T = Trimming I = IUPAC Basecalls M = Vector masking Chargeable service options S = Second pass in case of failure V = Base call verification H = Difficult structure, e.g. hairpin D = Difficult template B = BAC protocol
5 Warning: To Review Your Excel File Prior to Upload Click Check Order and click OK. If errors need to be corrected return to the my sequence tab and edit where required. Saving your Excel File Save as the Excel file on your computer Name the Excel file Close the Excel file Uploading the Excel File to the MyProject Web Portal In My new order in the dashboard, click directly on Upload, then browse to choose your file. Once the correct file has been selected, click on Submit to complete the upload Click on Upload Click Browse to find your saved Excel file Click Submit Once the file is uploaded, different coloured message will appear: GREEN The file was successfully uploaded
6 YELLOW (warning): Advises that the information entered for concentration, primer, etc. does not match GENEWIZ recommendations. You can choose to continue with your order or to delete your draft order, correct the errors as stated and re-upload. YELLOW WITH RED WRITING (warning with errors): The file has been uploaded partially but some lines were removed because they contained errors. Delete your draft order, correct the errors as stated and re-upload. RED (errors): The file contained too many errors to be uploaded. Correct the errors and re-upload the file. Notes If a second attempt is required, correct the errors as stated and re-upload, or contact Customer Services for assistance at DNASeqUK@genewiz or +44 (0)
7 Completing and Validating your Order As soon as the file is uploaded successfully click the circle against the reference of your draft order then click Complete order By default, sequence data is downloadable from the dashboard. If you wish to receive data by , please indicate the format Add additional comments if needed Save your order and Submit it Check all the details of your order and print your Production sheet When your order is validated and completed, you will find it under My current orders in the Dashboard In order to process the order, enclose the printed Production sheet with your sample shipment For more information please contact Customer Service at Tel: +44 (0) or DNASeqUK@genewiz.com
