1 Scepter Software Pro Quick Guide Scepter Software Pro is used to connect with one or more Scepter devices, upload test files for storage, perform data analysis, and create experimental graphs and reports. For a more detailed guide to the software, go to The Five Main Components of Scepter Software Pro Click Y-axis to adjust scale Use the checkboxes to choose files for the Multifunction Plot Click X-axis name to toggle between Diameter and Volume Choose Acquisition to view stats for Current Acquisition Choose Overlay to view stats for Multifunction Plot 1 Data Panel Displays all data sets used during an analysis session and provides access to previously saved data as well as files directly uploaded from a connected Scepter device. Each data folder for Scepter is displayed as a major node (e.g., Device SN 80094SL) with contained data files beneath. Use the icons at the bottom of the data panel to: 1A. Open data folders 1B. Save changes to data files (e.g., added or repositioned markers) 1C. Remove nodes (data folders) from the current data panel

2 2 Analysis Template Houses a set of user-defined histogram overlay marker(s) to analyze one or more test files in the Multifunction Plot. Only one template can be open at a time. Use the icons at the bottom of the data panel to: 2A. Open templates 2B. Save templates 3 Current Acquisition Plot Displays data from any test file highlighted in the Data panel. The plot can display data in either diameter or volume modes (X-axis) and at scalable cell counts (Y-axis). Statistics are instantly updated in the Acquisition table. Changes are not permanent but can overwrite the current file or be saved as new tests using. Use the icons at the top right of the data panel to: 3A. Expand the plot size 3B. Add/remove markers 4 Multifunction Plot Uses overlays to simultaneously display and analyze data from multiple test files with newly defined markers. Statistics derived from this plot are displayed in the Overlay table. A setup feature allows customizing of histogram color and other visual properties. Use the icons at the top right of the data panel to: 4A. Expand the plot size 4B. Add/remove markers 4C. Customize the overlay appearance 5 Group Stats Displays statistics and conditions associated with test run(s) in either the Current Acquisition Plot or the Multifunction Plot. To display data from the Current Acquisition, choose Acquisition. To display data from the Multifunction Plot, choose Overlay. The Setup and Notes/Comments features can be used to customize reports. Results can be exported for further analysis or printed as concise reports. Connecting to the Scepter Device and Uploading Test Files To connect to Scepter Software Pro, turn on the Scepter device and attach it to the computer using the USB cable provided. Click the Scepter application shortcut on your desktop. After the program launches, use the dialog box to connect to Scepter devices as well as upload files. Scepter Software Pro Quick Guide 2

3 Uncheck if you do not wish to save files Click to choose a folder location for upload You can also connect a device and upload files through the Device Panel icon or the Tools menu. More than one device can be connected Set Date/Time Access Sensor settings Once connected, the name of the device (DEVICE-Scepter SN) appears in the Data panel, with the uploaded test files listed below the device name. Test 01 is highlighted and its data is displayed in both the Current Acquisition and Multifunction Plots. Statistics for the entire data set are shown in the Group Stats window, in the Acquisition table. Scepter Software Pro Quick Guide 3

4 NOTE: If the Scepter device is asleep or if it is accessing histograms, settings, or help screens, Scepter Software Pro displays an error message, and the connection is aborted. To access previously uploaded test files, click the Open icon below the Data panel or click the File menu and Open. Use the Browse Folder dialog to select a folder containing saved test files. To remove folders from the Data Panel, click the Remove icon at the bottom of the panel. NOTE: Removing folders from the Data panel does not delete folders or test files previously uploaded to the computer disc. To disconnect, click Disconnect on the Scepter device screen or unplug the device from the USB port. A dialog box will remind you to save any unsaved changes. Analyzing Individual Test Files Using Current Acquisition Plot To analyze individual test files, click any test file in the Data panel or Group Stats table to display data. For newly uploaded files, the histogram marker (M1) is positioned as it was originally acquired on the Scepter device. To change the file displayed, click a different test file. To adjust marker positions: Vertical adjustment Click and hold on the marker region and then move the bar up or down. Horizontal adjustment Click and hold on either end of the marker to expand or narrow the region. NOTE: You cannot grab and move the marker. To add or remove markers, use the icons. For each file, new markers are numbered in order (as M2, M3, and M4 ). For each new marker added, a set of statistics appears in the Stats table for that file. Once a marker is adjusted or a new marker added, the Stats table updates all statistics to reflect the changes. To enlarge the plot, click the Zoom icon. To toggle between cell diameter and cell volume formats, click on the name for the X-axis (Diameter or Volume). To access a menu of pre-determined cell count ranges, click on the Y-axis, or click on Custom Count. Scepter Software Pro Quick Guide 4

5 Saving Changes to Data Files Adjustments to the markers (move, add, remove) in test files are not permanent; edited test files must be saved before quitting the application. To save changes to a single file, click on the test file to highlight it and then click the Save Data File(s) icon at the base of the Data panel. Or, select the File menu and Save As. Choose a folder and if desired, rename the test file. Saving the edited file in the same folder as the original causes the original file to be overwritten. Alternatively, click on the File menu and select Save to directly overwrite a test file. To save changes to multiple test files, hold down the Ctrl key and click on multiple files to highlight them. Click on the File menu, and then click Save. Or, click on the Save Data File(s) icon and then Save As. Select a folder, then from the dialog box, choose to Overwrite original files or Save as Copy. Copies of each file are named as Copy of file_original Name. Files cannot be renamed when multiple files are saved simultaneously. Analyzing Multiple Test Files The Multifunction Plot creates overlays from multiple data files. This plot has all the same features as the Current Acquisition Plot. Statistics from the overlay plot are displayed in Groups Stats in the Overlay table. To display this table, click the Overlay button at the top of the Group Stats window. NOTES: Files located in different data folders or Scepter nodes cannot be overlaid. If you wish to overlay test files located in different folders, save copies of the files to a new folder. To select files for display in the Multifunction plot, check the boxes to the left of all the test files in the Data panel that you want to include in the overlay. Selected test files lose their markers (M1, M2, and M3) but the multifunction plot allows you to create a new set of markers, which are applied to all test files in the overlay. To add/delete or adjust markers, follow the same procedure as in the Current Acquisition Plot. New Multifunction plot markers are labeled O1, O2, O3, etc. The Analysis Template is a set of markers (found under Region List) and statistics used to define a particular set of events. When Scepter Software Pro is launched, the Analysis Template panel displays a single template named Default Template. To enter a new name, double click on the default. As histogram regions are added, each is listed in the Region List under the template name (O1, O2, O3 ). To save or recall an Analysis Template, use the Save Template or Open Template icons at the bottom of the Analysis Template panel. NOTE: Only one Analysis Template can be open at a time. Therefore, opening a saved template replaces the template that was previously housed in the panel. Any changes made to the previous template are lost if the changes were not saved prior to replacing it. Scepter Software Pro Quick Guide 5

6 To adjust the appearance of the Multifunction Plot, access the Overlay Setup feature through the Overlay Setup List icon. NOTE: Features associated with Overlay Setup are slightly different in the Macintosh version of Scepter Software Pro. Dilution Calculator Scepter Software Pro can be used to automatically calculate original cell culture concentration and cell counts based on statistics for diluted samples. The calculation is based on the values entered in two columns in the Acquisition table Dilution Factor and Original Volume. Dilution Factor is the amount the acquired sample is diluted relative to the starting sample. Original Volume is the volume, in ml, of the starting sample. The default value for both is To calculate the original concentration and cell count, check the boxes above the columns Dilution Factor and Original Volume in the Group Stats Setup window. Then, double click on the default value (1.00), enter the correct value, and click Done. The software recalculates statistics for cell counts and/or the concentration for the total events and all marker-defined subsets. NOTE: Changes to the Dilution Factor affect both concentration and cell count values. Changes made to the Original Volume only affect the cell count values. Formatting and Printing Histogram Plots and Statistics All statistics are presented in the Group Stats window in two tables, Acquisition and Overlay. The Acquisition table shows stats for the highlighted test file along with all test files that are listed in its data node. Stats for test files from other data nodes or Scepter nodes are not included in the table. The data includes original stats acquired by the Scepter device as well as any additional markers applied after uploading. Data from the Multifunction Plot is displayed in the Overlay table. This table contains only the test files and markers used in the Multifunction plot. Scepter Software Pro Quick Guide 6

7 To toggle between the two display formats, use the Acquisition and Overlay buttons at the top of the Group Stats window. To customize fields for a tabular report, use the Setup button at the bottom of the window. Use the Notes/Comments field to summarize the experiment. Summaries are included in both the exported CSV file and the printed report. Uncheck any column to remove it from display Uncheck Show to remove a test file from display Use the Sample column for naming test samples Click Done to exit The stats tables can be exported as independent csv files or directly printed. To export as a csv file, use the Export to CSV button at the bottom of the Group Stats panel. To directly print a stats table, use the Print Stats button at the bottom of the Group Stats panel. Use the Printout Preferences dialog box to format the printout, then click Done. The printout includes the current date and the entire table as displayed in the Group Stats window. The printout for the Overlay table also includes the associated Multifunction Plot and a color-coded legend.
