2 Sensitive Epi-Fluorescent Imaging for Assay Development & High Throughput Screening The ImageTrak is an innovative system that integrates fiber optic epi-fluorescence imaging with PlateTrak automated liquid handling robotics to provide a high capacity, high throughput system for assay miniaturization and lead optimization. The ImageTrak is unique in its ability to combine proven Packard automation, sensitive imaging techniques and data analysis features into one complete system for high throughput screening. Two ImageTrak models are available to handle varied assay requirements. Each system is equipped with 96-channel dispensing for automated processing of 96 to 1536-well high-density microplates, a bi-directional conveyor for secure plate handling and an epi-fluorescence imaging module in a light tight enclosure. The ImageTrak utilizes 96 individual fiber optic bundles with separate optical paths for excitation and emission light to enable simultaneous measurement of 96-wells in dual wavelengths for true ratiometric FRET and kinetic assays. The ImageTrak s optical system is mounted from below and built into Packard s unique MultiPosition Dispense (MPD) module design. The MPD combines Dispense Head movement, tip and plate handling, and liquid dispensing into a single unit for precise automation control. Using the MPD, the ImageTrak can accurately position microplates on the contact imager with tips loaded, enabling simultaneous dispense and read capabilities. The ImageTrak HT model offers an additional 10-inch MPD with 384-channel Dispense Head to pre-load dyes and dispense compounds for high throughput liquid handling. System Features > The ImageTrak provides automated control of dye loading, reagent dispensing, tip changing, plate washing, timed incubations and compound, agonist and antagonist additions for automated assay screening. > The ImageTrak Epi-Fluorescence detector utilizes separate optical paths for excitation and emission light at two wavelengths to minimize cross talk between channels (<1% in 1536-well plate). > ImageTrak provides simultaneous dispense and read capabilities using a 96-tip Dispense Head when reading from below. > Contact Imaging with fiber optics throughout the system for high light gathering efficiency. Fiber optics eliminate the need for complicated lenses, avoiding lens effects such as vignetting, reflections, cross-talk, and removes the requirement for focusing. ImageTrak MultiPosition Dispense (MPD) module holds reagents for liquid handling, a 96-tip Dispense Head with integrated Gripper, and a positioner for accurate plate placement on the Fiber Optic Imager.

3 The ImageTrak is an innovative system that integrates fiber optic epi-fluorescence imaging with PlateTrak automated liquid handling Dispense Head P50 Dispense Head enables versatile 96- channel pipetting with automatic tip loading from the conveyor path or labware deck. The ImageTrak HT system includes an additional P tip Dispense Head for pre-loading of dyes prior to positioning on the contact imager for dispense and read. MultiPosition Dispense Module 30-inch MultiPosition Dispense (MPD) module with 96-channel Dispense Head and Integrated Gripper. The MPD is capable of variable positioning in the X, Y and Z-axis with 0.01mm resolution. The MPD includes a plate positioner over the CCD camera and nine deck positions for accessories such as tip wash cartridges, recirculating chillers, reagent bottles and more. The ImageTrak HT system includes an additional 10-inch MPD and 384 channel Dispense Head. MPD Gripper Integrated MPD Gripper tool for moving microplates from the conveyor path onto the plate positioner and onto other deck positions. Enables plate positioning with tips loaded for increased speed and efficiency. Bi-Directional Conveyor Bi-directional conveyor for microplate and tip movement. Conveyor and MPD can operate simultaneously for true parallel processing. Robotic Shuttle Robotic shuttle for access into the light tight enclosure and for integration with external devices using the Packard SideTrak TM for placement into incubators, washers and more. Contact Imaging CCD Camera Patented "contact imaging" with cooled CCD camera and 96 fiber optic bundle untilizing separate optical paths for excitation and emission light to measure 96-wells simultaneously. Xenon lamp excitation light source is provided as standard for variable excitation wavelengths from nm. Excitation light is distributed efficiently by means of fiber optics to each of the 96-sites independently. Emission wavelengths range from nm with a 6 position filter wheel for choice of 3 emission pairs. Universal Stacker Modules The ImageTrak holds three Universal Stacker II modules, each with capacity up to 50 SBSstandard microplates, 15 deep-well plates or Packard Disposable Tip Boxes. Stackers can be removed and replaced for increased system time and capacity. The ImageTrak HT uses two Stack Doubler modules, each holding two Stacker cassettes.
