3 SI8000 Cell Motion Imaging System The Sony SI8000 Cell Motion System uses high speed video microscopy to detect and record cell motion. A unique motion vector analysis algorithm quantifies cell motion. This technology permits monitoring of non-labeled cells over time in their physiological environment so that cell motion can be analyzed in a non- invasive manner. The system leverages Sony expertise and patented imaging technologies for the ultimate sensitivity, speed, and resolution of cell motion. In addition, the SI8000 uses vector analysis originally developed for video decoding to analyze cell movement. The Sony SI8000 won the 2015 outstanding new product award from the Science and Laboratory Automation and Screening (SLAS) organization. 4 High Speed Motion Capture, Quantification and Visualization The Sony SI8000 system is equipped with a phase contrast microscope, a high-speed camera, CO2-incubator and specialized software. The system automatically detects and adjusts image focus and light conditions. Video imaging captures activity at a high frame rate (150 fps) to deliver data rich results for analysis. Motion vector analysis, based on video analysis technologies developed by Sony for its audio-visual products now allows researchers to analyze movement of cells over a period of time. This permits the system to be effective for analysis of single cells, cell membrane, a single monolayer of cells or for cell-to-cell interaction and intracellular activity. 5 The SI8000 is label-free so that cells may be observed in their natural state ensuring that there are no outside interactions from potentially toxic dyes or fluorochromes in the environment. This capability is particularly useful for toxicity studies as it allows researchers to see how cells behave as a drug is added to the system. An optional CO2 incubator reliably manages an optimal environmental for in-vitro cell observation. The scanning stage controls the X, Y and Z position of the imaging system for precise and repeatable positioning. Data analysis software lets researchers quantify and visualize cells in motion. A variety of contractile parameters are supported by quantification including contraction and relaxation speed, average deformation distance as well as contraction and relaxation duration. Power Spectrum Density, that measures signal strength is also supported. Graphic outputs include heat maps of contraction, relaxation, propagation and isochrone maps used to detect abnormalities in cardiac cells. Cellular and sub-cellular motion visualization is also supported. Quantification Quantification capabilities includes contraction speed, relaxation speed, average deformation distance, contraction duration, and relaxation duration. Power Spectrum Density measures signal strength. Visualization Visualizations include heat maps of contraction and relaxation, propagation of contraction and isochrone maps used in cardiology to visually detect abnormalities.
