3 Introduction Thanks for your purchase of the Persee Analytics Inc. T3 spectrophotometer. UV-Visible Spectrophotometer is a well-accepted, documented technique with many applications. The technique is extensively used for the analysis of agricultural products and is widely used in public health, environmental protection, life sciences industries and many other organic and biochemical applications. For obtaining maximum performance and trouble-free operation, we give you the following suggestions: Upon receiving your instrument, please check all the items listed in the packing list are included and install the instrument according to the instruction manual. The spectrophotometer is a precise electronic instrument. Please 1

4 read the instruction manual carefully before operation. If the equipment is used in a manner not specified by the manufacturer, the protection provided by the equipment may be impaired. Avoid collision and handle the instrument carefully during transportation. If the tested solution is harmful to humans, please wear gloves or use other protection methods. Please keep the accessories in a safe place for the use in future maintenance. For more information, please visit 2

5 Notice: The light source of the instrument must be preheated for 10 minutes. It can start measurement after preheating. Please ensure that the battery is charged amply before using the instrument. The charging time should not be too long. Using the touch pen to operate on the touch screen, avoid using edge tool to destroy the touch screen, avoid pressing the touch screen forcibly. Avoid blocking the exhaust fan, avoid working in the high temperature environment. In order to avoid destroying the instrument, strong impact should be avoided. Instrument should be placed in a stable working platform. Strong vibration source should be avoided during the measurement. Take some safety measure arrest the thunder when installing antenna in the wild. It s forbidden to absorb the magnet of the antenna on the object with electricity. Instrument should not work in that environment where the humidity is higher than 85%. 3

10 Chapter 1 Introduction 1.1 Uses T3 is a small portable quick-test instrument, whose measurement speed is very fast. The dimension of the instrument is small, and it s light. It is convenient for carrying and manually operation. It can not only be used in the lab, but also special for the wild environment monitor and on-site real time detection. The instrument is provides with the function of spectrum scanning, kinetics, quantitative measurement and photometric measurement, so the application field of it is so wide. The measuring method is based on the Lamber-Beer s Law, which is found on the absorption to light of the material. 1.2 Structures The T3 system is consist of main unit, cell holders, test tube holders and fiber probe. 1.3 Characters ARM11 high-speed embedded processor and embedded real-time operational system WINCE6.0 are used for the core control part of the T3, which provides 8

11 perfect software and hardware platform for the functions implementations of the instrument. For the convenience of user's operation, extra large display touch-screen is used for display. The monochrometer adopts concave plate holographic grating made by French JY company integrated with CCD ILX511 detector made by Japanese Sony Company featuring with high wavelength accuracy and low stray light. The light source adopts gathering micro tungsten lamp with the characteristics of small volume, stable illumination and long life time, which can meet the special requirements for portable devices. 1.4 Functions and configuration Spectrum Scanning, Photometric Measurement, Kinetics, food analysis, water analysis, video player and other user-defined functions (quantitative measurement). The main unit can be operated stand alone, save and read the data and print directly. High-Lattice touch screen. It can be operated by touch pen or mouse. Communicate by the USB and RS232 port, LAN communication is reserved. Equipped with GPRS/GPS module. Support video play function. 9

12 The optical structure support multi kinds of sampler holders changeable and the light source energy can be adjust automatically. Built-in chargeable battery, it can work for 5 hours with one charging. (with full loaded, it can do precision measurement for 4-5 hours ) Configure with V/50Hz-60Hz charging power supply, it can offer power for the main unit and charge the battery. Offering micro printer. 15 There are two kinds of test tube holder: 1. φ25 test tube holder: shown as figure 2-2. Loose the fixed screws off the hole, insert theφ25 test tube into the test tube holder, and the test tube is fixed by the spring. Insert theφ16 test tube into the adapter, and the test tube is fixed by the spring. 13

17 2.3 Cell holder accessory Shown as figure 2-5. T3 / T3M Instruction Manual Figure 2-4 Cell holder The cell holder is consist of holder, baffle and spring. It supports 10mm, 20mm, 50mm cuvettes. The baffle and spring work together to fix the position of cuvettes, assure the precision of measurement. Installation: 1. Put the cell holder into the sample compartment. 2. Tighten the fixed screw. 3. Insert the baffle and spring into the cell holder as the cuvettes dimensions. 4. Clean the cuvettes after finishing measurement. 5. Loose the fixed screw, and remove the cell holder. 15

AC Plug: Connecting with the charging power supply, power for the main unit and charge the battery. Charging indicator: The indicator is lit when charging the main unit, it is off 17

20 when the charging is finished. RS232: The interface is connected with printer and accessories through special communication cable. USB main interface: Support the flash memory, saving the data into the flash memory. Also can connect with the USB printer. USB slave interface: Connect with the PC. Download, view, export and print the data by the special software T3 data viewer. SIM card: Insert the SIM card for GPRS communication. Here is the insert method of the SIM card as figure 2-8 shown. Figure 2-8 SIM card insert method LAN: Connect with the LAN for internet communication. GPS interface: Connect with the GPS antenna to acquire GPS data. 18

21 2.5 Connection between the main unit and PC/Accessories The RS232 interface is connected as Figure 2-9. One side of the special communication cable is connected with the RS232 port, while the other side is connected with the printer. Print the data and spectrum directly by pressing Print button on different modules. The communication method between the T3 and PC is the same as that mentioned above. One side of the USB cable connected with the USB slave interface, while the other side is connected with the PC, the communication will be built after synchronization. 19

24 Figure 2-11 Power display There will be some notice if battery is low. See figure Please use the charging power supply for offering power to the main unit and charging the battery. The charging power supply should be connected with AC of V/50-60Hz. The other end is connected with the charging interface on the main unit. The power supply indicator is lit. The screen displays charging. After the charging is finished, the power supply indicator will be off. The screen displays. The standard charging time is 5-6 hours. The 22

25 recommended charging time should not be too long to avoid damage the battery. The charger specification is V/50-60Hz input and 18.5V/3.5A output. Figure 2-10 Low battery Chapter 3 Measurement accessories instruction Note: The flexing diameter of the fiber should be bigger than 0.1 meter to avoid the inner fiber breaking. Fix the fiber probe in the holder when measuring, and keep it not moving. 3.1 Fiber probe This instrument takes fiber probe to take photometric measurement. Put the immerging fiber probe into the sample solution to measure. Here are the 23

26 specifications of fiber probe: Wavelength range: nm Pathlength: 10mm, 20mm (interchangeable) Standard SMA 905 port Attached with holder, easy to install on site. The reflection mirror can be disassemble, easy to clean The material of the fiber probe is titanium alloy, which is acid and organic solvent resistant. Figure 3-1 Fiber probe assembly 24

27 3.2 Using method The branching ends of the fiber probe connect with the light source export and sample light import respectively, choose different path length according to different measure method. Insert the fiber probe into holder. Measurement: choose the corresponding method, put the fiber probe into a blank solution to do zero correction and sample to measure. Clean the probe with a filter paper and wash with the solution to be test three times. Note: If there is bubble existing on the top of the probe after inserting into the solution, please remove the tested solution and shake the handle gently. The measurement can t be done until the bubble is removed 25

28 Chapter 4 Operation 4.1 Instruction before using Interface introduction There are four parts of the interface, which are status bar, navigation bar, current page and tool bar. See figure 4-1. Status bar Navigation bar Current page Tool bar Figure 4-1 Interface Status bar: Shown on the left of the status bar are the current measurement mode and flash memory notice, the choices are transmittance (T%), Abs and Energy (Es), on the right is the current time and battery status. 26

29 Current page: The current page is switched by the navigation bar. Navigation bar: In the middle of navigation bar is the current measurement, shown as figure 4-1 is Spectrum scanning. The button on the both sides can be used to switch between different pages. Go back to the measurement page by pressing button. Only on measurement page can switch to the other navigation pages. Tool bar: List the operation that can be carried on the Current page. The chosen items character will be bold. Taskbar: A dynamic window pop-up when a certain task is performed, the window automatically disappears after the task is completed. Click to abort the task and hide the window. There are also the buttons as follows, the Figures below are only for examples, the contents displayed will change in application. Multiple choice button: 27

30 Click the option you want to select, the background turns blue. If the color doesn t change after clicked, then it means you cannot select this option. Multiple choice drop-down box: Click the drop-down arrow, the following option box, click the option you need. Input & edit box: If you want to change the content in the input box, click the content on the right, the program will automatically detect the content input, and a corresponding number or text input panel will pop up for user to edit. 34 Translate; Full screen; Read screen point data; Change the range of coordinates; Distance measuring, positioning by dragging the acute angle of the triangle; Read the point value on the spectrum, if click this button, use the arrow moving from left to right to select the date point; Select the area of interest; Move the whole processing panel; Exit the spectrum processing. 32

35 4.1.2 Auto Zero Before measuring the transmittance (T%) and absorbance (Abs), the use needs to carry out the zero calibration. The steps are as follows: Step one: put in the blank solution, press the button <zero>, which reminds you to put in the blank solution, Figure 4-6 Then press the button <OK>, the following taskbar will pop up: Figure 4-7 Step two: if the Auto Zero is successful, the taskbar will disappear automatically Sample Measurement After the auto aero is finished, put in the solution you want test, click <measure> on the main page, a taskbar <measuring> will pop up. Click to abort the measurement. The taskbar will hide automatically when the measurement is finished. 33

36 The process of auto zero and measurement will not be mentioned again in the following description. 4.2 Operation Main Interface Switch on the mainframe, the main interface (see Figure 4-8) will display after the boot screen. This unit is touch screen control, press the key areas with touch pen or mouse 34

37 will perform the corresponding functions. The following is the introduction to the six functions Food analysis This module includes more than 50 methods of food analysis: nitrite, sulfur dioxide, formaldehyde, cyanide and peroxide value, etc. User can also customize dedicated test methods (under "quantitative measurement" mode). Ordinary procedure of <food safety>: select measuring methods-select measuring module-measure sample-save data-check data-print data. (Refer to<instruction manual of food safety analysis> for more information). Fig4-9 is the method-selection interface of <food safety>. 35

40 Quantity: input the mass or volume of sample. Dil. Factor: If the concentration exceeds the concentration limit, it can be diluted before the test, as a baseline for measurement. Click the on the navigation bar to set the parameters. The interface as Figure 4-12 will pop up, which consist of parameter option page and instrument option page. 48 Figure 4-20 Reference: measure the reference solution, then get the value A1 of reference solution, timer begin to count down. Sample: measure the sample solution, then get the value A1 of sample solution. Put the reference solution into test tube holder when the timer is to finish counting down, the value of A2 of reference solution will be automatically read out. Put the sample solution into test tube holder, then the value of A2 and inhibition ratio will be obtained. 46

49 4.2.3 Water quality analysis This module includes more than 30 methods of water quality analysis: Cr VI, cyanide, free chlorine and sodium reagent ammonia, etc. User can also customize dedicated measuring methods (under <quantitative measurement> mode) The ordinary procedure of <water quality analysis>: select measuring methods-select measuring module-measuring sample-save data-check data-print data (Refer to <instruction manual of water quality analysis> for more information). 62 The options can be set up are: Single wavelength/dual wavelength: set the measurement wavelength 1 st Order/ 2 nd order: times of fitting Coefficient a, b, c: in order, coefficient of constant, the first degree polynomial, and the quadratic degree polynomial. Concentration range: display the upper and lower limits of concentration of this method. Timers Count: modify the number of timers and the time for them. Significant figure: number of significant digits displayed. Unit: Display or set the unit of the method. +ve Slope/-ve Slope: display whether to choose fading measurement or not, among which the values of absorbance, slope, and correlation coefficient are all negative (COD low concentration). 60

Delete all: clear the list of all the methods. Note: if the user does not select the method, when the quantitative measurement is opened, the default parameters will be those of previous measurement Photometric Measurement Photometric Measurement means to read the measuring data at the certain wavelength. That is to read the data at a fixed point, the user can decide a 68

71 wavelength point and do the photometric measurement. The process is: set measuring parameters measure samples save data view data print data. Figure 4-42 See Figure 4-42 for photometric measurement interface. Press button into setting page, see Figure

102 Chapter 5 T3 Data Viewer Installation and Operation T3 data viewer software can be run under Windows2000/XP/Vista/Windows 7, which is used to communicate between PC and T3. The software can read the data from instrument, process data and print them out. T3 data viewer can read the data in the flash memory copied from T3 main unit. T3 data viewer can export the measured result or the spectrum to text document (.txt or.csv) T3 data viewer can print the measured result or the spectrum by the printer connected with PC. T3 data viewer can be used to update the instrument. 5.1 Software installation: T3 data viewer must be run on the PC with following software: Microsoft.net Framework 2.0 Windows Mobile Device center provides the device manager and data synchronization function between mobile device and PC. USB drivers used to recognize T3 main unit which is connected with PC by USB cable. 100

Double click the T3 data viewer v1.0 icon from the desktop to start. Start-Program-T3 data viewer v1.0 Double click the T3 data viewer.exe from the installed file. There are four parts for the software interface: menu, toolbar, document list, data browsing page. There are six functions of the data browsing page: spectrum, kinetics, quantitative measurement, food analysis, water quality analysis, photometric measurement, which are used to browse and process the data with corresponding type. Display the calibration curve, parameters and measurement result on Quantitative page. Also can be printed and saved. When open several measurement data with the same calibration method, new lines will be add to the measurement result list. When open with different calibration method, the measurement result list will be displayed from the first line. When open multi files, stop browsing or printing by choosing Edit - Delete current Row s File Data. The interface is shown like this: 112

124 Chapter 6 Instrument maintenance 6.1 Maintenance cautions The main unit and accessories should be put into a convenient box and be locked for transportation. The immerging probe should be cleaned with distilled water and be covered with a rubber cap after the operation. After long-time use, the inner surface of the probe maybe has some filth. Remove the tip of the probe and clean it with a alcoholic cotton tack. It should avoid dust covering the surface of the reflecting mirror after the fiber probe is used. If the operation is not done for a long time, please turn off the power supply of the lamp to save the battery energy. After the charging is finished (the indicator is off), please turn off the power supply, to ensure the battery life and the safety of the instrument. Temperature range: operation environment is 5-30 and store 122

125 environment is Replacement of battery The effective charging times of the rechargeable batteries inside the instrument are The procedure of the replacement of batteries (purchasable from the manufacturer) is as follows: 1. The trouble phenomenon is that the energy scanning reading is close to zero when the lamp is lit. 124

127 Replacement for fiber detector top: After the fiber probe has been used for a long time, the energy is lowered evidently. The trouble phenomenon is that the energy reading at 560nm is lower than 10 when the gain is 1 and integrating time is 0.005s in energy scanning. Please contact the manufacturer for replacement. The replacement methods are as follows: The valid time for the fiber detector top is 2 years. When the energy is lower, please contact the manufacturer for replacement.
