5 Testing a Sample Sample Preparation Acceptable sample types: Serum, Lithium-Heparinised Whole blood or Plasma Volume Required: The system uses 200ul per test panel Testing must be conducted within 60 minutes (at room temperature) To prevent haemolysis of the sample, do not refrigerate, freeze, or shake whole-blood sample. Plasma or serum can be stored at Room Temp for 5 hours prior to testing or up to 48 hours if stored at 2-8C Performing the Test Turn the analyser on (and off) by pressing the power on button on the front panel. The system automatically launches the warming procedure and completes the system self test. This may take up to 15 minutes for the system to become ready depending on the current room temperature. Preparing the Disc Discs should be stored at 2-8C and can be used straight from the refrigerator without the need for warming - Avoid placing unopened reagent discs in temperatures greater than 25C (77F) for more than 48 hours. 1. Before using a reagent disc, carefully check the foil pouch for any damage. Tear open the package from the upper right side of the pouch. 2. Remove the reagent disc gently with finger and thumb along with the tissue paper from the foil pouch. 3. After removal use your other hand to hold the disc by the edge. Avoid contact with disc surface where optical measurement takes place. 4. Unwrap and discard the tissue paper. 5. Check the reagent disc for any damage do not use reagent discs that have been dropped or damaged. 6. After opening the pouch the disc must be used within 20 minutes. Do not place the disc back in the refrigerator for later use. 7. Holding the disc by the edge, gently pull the end of the aluminium strip in an outward direction along the surface of the reagent disc; this will reveal the sample port. Discard the aluminium strip. 5 VB1 DM/JAC 2014 v1.0
6 Dispensing the sample 1. Attach a new pipette tip to the end of the 200ul pipette provided. Do not touch the end of the tip. 2. Using your thump push down the plunger on the top of the pipette to the stop position. 3. Immerse the pipette tip below the surface of the sample. 4. Slowly release the plunger to draw up the sample, avoid drawing in any air, as this will cause the disc to fail. 5. Remove the pipette from the sample tube. 6. Insert the pipette tip into the sample port. The pipette tip should only make slight contact with the bottom of the sample chamber, do not push down hard. 7. Keep the reagent disc level and the pipette tip perpendicular to the surface of the disc. 8. Gently push down the plunger at the top of the pipette to the stop position. All of the sample should have been expelled into the sample chamber. Keeping the plunger held down gently remove the pipette tip from the sample port. The plunger may now be released, remove the disposable tip from the pipette and discard the tip. 9. Press the START icon on the touch screen to open the reagent disc drawer at the front of the analyzer. 10. Hold a reagent disc that contains a sample by its edge, with the barcode side facing up and the disc kept level. Avoid touching the surface of the reagent disc. Gently place the disc into the drawer and press OK to close the disc drawer. 11. Use the touch screen to enter the patient ID (up to 12 numbers) or scan the patient ID using the optional external barcode scanner and press OK 12. If you press OK without entering a patient ID, the system will use the current date and time as the patient ID in the format dd/mm/yyyy hh:mm. 6 VB1 DM/JAC 2014 v1.0
7 13. Select the species (feline, canine, and others). 14. The fully automated analysis will now commence once the disc drawer closes 15. When the analysis is completed, the system will store the analysis results and display them on the screen. To cancel a test in progress, press the CANCEL icon on the screen, the system will prompt for confirmation. A reagent disc cannot be re used if a test has been cancelled. Test Results When the test is completed the system will display the results produced. Press the Printer icon to output the results via the built in thermal printer. Press the Home icon to return to the main menu and press OK to open the draw and remove the used reagent disc. 7 VB1 DM/JAC 2014 v1.0
8 Why Run a QC? How to Reconstitute a Biochemistry Control Vial -To assess the quality and performance of the practice laboratory. -To ensure that results generated by the Skyla VB1 are correct. -QC monitors staff, equipment, reagents and result reporting. Preparing the QC reagent 1. Remove vial from the refrigerator and allow to warm at room temperature for 5 minutes 2. Carefully open and remove the bung from a vial of serum and a vial of serum diluent. 3. While holding the serum vial steady on the bench, carefully pour all the diluent into the serum vial. Do not shake excess diluent out of the diluent vial, as this has been accounted for in the dispense. 4. Close the serum vial, swirly gently -DO NOT shake. Let the vial stand on bench for 20 minutes then swirl again and gently invert 10 times. 5. The control is now reconstituted and ready to be analysed or frozen for prolonged storage. QC Storage: Store at 2-8 o C before and after reconstitution. Reconstituted control may be stored at -20 o C. Thaw frozen controls quickly using warm water (37 o C), invert gently after thawing. QC Stability: Open vial reconstituted stability is up to 10 days if stored at 2-8 o C, or 2-3 weeks if stored at - 20 o C once reconstituted. Frozen controls must be used within 8 hours of thawing. 8 VB1 DM/JAC 2014 v1.0
9 Running a Quality Control 1). Using the prepared QC sample follow the procedure as indicated in Testing a Sample for dispensing the sample. 2). Press SETTINGS on the main menu screen. 3). Press the CONTROL icon. The test process is the same as the one for sample analysis; however there is no need to input any patient related information. 4). The QC control test results are stored separately from sample results. The results can be printed out after the test is completed. 5). Past QC results can be accessed using the RECALL function (See the Skyla VB1 Operator s Manual for more details. 10 Interpreting the results The printed report consists of two sections. The heading at the top of the report includes information about the analyser model, test date and time, sample type, patient ID, reagent disc serial number, species, gender and weight. The results section consists of four columns: Test marker, analyte concentration, ref range and unit. The test report also shows the degree of Lipaemia (LIP), Haemolysis (HEM) and icterus (ICT) of the sample analysed. Symbols Used in the test Reports: (#) Calculated value (*) Test result outside the reference range (%%) The test results are significantly impacted by the interference of LIP, HEM or ICT of the sample that is outside the permissible range of 20% >or< Indicates that the result is outside the measurable range of the analyser. The value displayed is either the lowest or highest value that the analyser is capable of measuring. For Example, the measurable range of glucose is mg/dL.. If the result is less than this range, <70 mg/dl will be shown or higher that this range, >600 mg/dl will be shown. (+) indicates the degree of Lipaemia (LIP), Haemolysis (HEM) and Icterus (ICT of the sample. The more + symbols then the more severe the interference. The classification is as follows: 0 = Clear or without interference + = Mild ++ = Moderate +++ = Severe NA indicates that the result cannot be calculated 10 VB1 DM/JAC 2014 v1.0
11 Accessing Previous Results The Skyla VB1 automatically stores tests results for display, printing or Export to USB To access previous results: From the Home Screen > press <Recall> then <Search> > > Select the required search method and follow the instructions on the screen. 12 Loading the Printer Paper Open the top cover of the built-in thermal printer and place the printer paper into the paper slot. Confirm that the heat-sensitive side is facing the correct direction (the arrow sticker on the roll should point downwards). 12 VB1 DM/JAC 2014 v1.0
13 Date and Time setting The system date and time will usually be set before installation and the system remembers the date and time between uses. To Change the date and time: Select settings from the Home screen Select administrator settings Select Date & Time Select the Date/Month/Year Hour/ Minute and Use the Up and Down arrows to adjust the value. 14 Troubleshooting Common Problems Analyzer Power and Function C = Common problem S = Solution C: Touchscreen is black or blue; power light is OFF. S: Turn the analyzer power off. Check the power cable connections. Disconnect and reconnect on both ends, then turn the power on. C: Touchscreen is blue or blank; power light is ON. S: Remove the reagent disc from the drawer. Check that the drawer closes properly, then re-boot the analyzer. C: Damaged touchscreen S: Please contact the authorized distributor or technical support department. Settings and Reports C: There are no values for some of the results within a printed test report, or the system automatically cancels a reagent disc test without generating a report S: The skyla VB1Veterinary Clinical Chemistry Analyzer performs automatic system calibrations to maintain high system precision. Thus, when the Analyzer detects a faulty reagent disc, it automatically cancels the test and displays an operational error message. Please insert a new reagent disc to resume operations. C: The test report exported by the built-in thermal printer is blank. S: The thermal paper roll may have been installed in the wrong direction within the built-in thermal printer. Please remove the roll of thermal paper and confirm that the heat-sensitive side is facing the correct direction (the arrow sticker on the roll should point downwards) before replacing the roll in the built-in thermal printer. C: After choosing to initiate the System Default function, previously customized reference range settings have disappeared. S: Once the System Default function is initiated, the system will delete all custom settings and reference ranges. Once the command is executed, all settings will be reset to the default system settings. The settings that have been deleted cannot be recovered. However, none of the test results and calibration records will be deleted. Exporting data from the system to other devices C: Peripheral and media of the exported data. S: Dump to USB function: will export selected historical data to an external USB drive. RS232 Export function: will export selected historical data to an RS232 station. 14 VB1 DM/JAC 2014 v1.0
