This cycle takes approx. two minutes. 13. The amount of CBC s left in the Pack will be indicated on this screen with a percentage bar showing the percent of fluid left in the pack. (This amount should be at 100% after the change pack procedure performed above.) 14. Select one of the main menu buttons to leave the Reagent Replacement menu. Reagent Pack Handling Recommendations 1. Write the installation date on the pack to distinguish it from a new pack. 2. Make sure all 4 red caps have been removed before pack is inserted into unit. 3. Make sure there are no obstructions in the reagent pack receptacle area before inserting the pack into the unit. 4. Verify waste connector is firmly installed and tubing is not being pinched by the top cover or by the pack. 5. Check questionable new or used packs for fluid by placing the reagent pack on its side so three bottom ports are visible and press a pen or screw driver into each port to see if any fluid escapes (if pack still has fluid, it will rush out rather freely so be prepared to have some paper towels to catch the fluid). If no fluid is coming out of any of the three bottom ports the pack is used up. A "new" or "used" pack will feel and weigh the same. Burp Reagent Pack Make sure that you Burp your pack regularly. Remove the waste line at the top of the reagent pack and press a pen or screw driver on the top port to release any build up of air inside the waste port. The more the machine is used the more regularly you will have to burp the pack. 3

12 Flags * - following either WBC, RBC HCT or PLT -- system analyzed the sample 3 times but all 3 counts differed and were outside the system s precision limits (see below). Result should be verified by repeating the sample. $ - between the test result and the units -- 3 counts were made and 2 were within the system s precision limits (see below). The result can be accepted.! - next to the HGB result -- the difference between HGB blank done for this analysis and the HGB blank done for the previous analysis was outside the system s precision limits. Instrument provides a result based on the previous HGB blank. Result can be accepted. If flag persists for more than 3 consecutive times, see section in the scil Vet abc manual to run checkup procedure. H / L behind result: Result is higher / lower than the reference value. ---D or < printed instead of a result -- linearity range for that parameter has been exceeded (see below). Repeat using a 1:2 dilution with saline solution. Multiply your results by 3. MIC following PLT Flags -- microcytes present in the platelet measurement zone. Verify result with manual count. SCH following PLT Flags -- schistocytes or platelet aggregates present in the platelet measuring zone. Review slide before reporting results. SCL following PLT Flags -- small particles in the 2-3 fl zone. Reanalyze sample. If flag persists, perform an automatic cleaning cycle and repeat again. If flag still persists, perform manual platelet count. AG1 and AG2 Flags for big Leucocytes aggregates (> 1,5 % / 2,5 % of total Leucocytes). Mix sample properly and repeat measurement. 11

13 Required: Caution Quality Control One 2.5-mL vial of Minotrol, Blood Control. (Low, Normal or High) Quality Control Target Values must be loaded on the Vet ABC Plus for the selected control vial. The Blood Controls; Must be stored in a refrigerator between 2 and 8 degrees Celsius. Can never be frozen at anytime. Cannot be left out for more than 15 minutes. Should not be held in a hand while waiting for control results, may get too warm Do not use a Blood Rocker. Blood Control Preparation 1. Read this entire Quality Control Procedure. 2. Remove Blood Control Vial from the refrigerator and roll the blood control vial in your hands until the vial is at room temperature. 3. Invert the vial back and forth slowly, paying attention to the concentration of cells at the bottom of the vial. There should not be a concentration of cells at the bottom of the vial when the vial is properly mixed. DO NOT SHAKE 4. The Low control mixes fairly easily while the Normal and High controls contains more cells and requires a longer inversion period (3 minutes of slow inversion). 5. When running multiple Blood Controls, always invert the vial 20 times before each sample. Do not continue to roll the vial in your hands. **All blood control vials are stable until the printed expiration date if unopened and stored in a refrigerator. Quality Control Handling Recommendations 1. Write the date on the control vial and cover it with tape to prevent it from rubbing off when vial is first opened. 2. The control vial should be mixed for a minimum of 3 minutes prior to running a quality control test. 3. If the control vial has been dropped or frozen it will damage the cells and cause aberrant results and flags. (ex. $, *, MIC, etc...) 4. Clean the internal and external threads of the cap and vial to prevent the blood from drying and flaking off into the control vial. 5. If a control vial has been left out too long or is too warm it will distort the HGB result. 12

14 Once the control is opened it is good for 16 days in a refrigerator if proper handling is followed. Quality Control Procedure Press the Quality Control menu button on the right side of the screen to enter the menu Quality Control. The Quality Control menu is composed of three tabs: 1. If any parameter failed the QC cycle the results will be highlighted in red on the screen. The results of all analysis are stored in an internal database for quality control analysis. The analysis as well as the patient results can be displayed and printed again as needed. 17

19 Calibration The scil Vet ABC Plus should be calibrated every six months. Before Calibrating Perform a concentrated cleaning Run a Quality Control test three times in a row to verify the instrument is reading consistently before proceeding with the calibration procedure. The values can be out of range but must be consistent with each other. If you are unsure of your results please call technical support before proceeding with the calibration. You will need to be logged in as "scientist" to be able to run the calibration procedure. DO NOT remove the Calibration Fluid from the refrigerator until step 6 is complete and you understand the instructions. Do not follow the instructions that come with the calibrator. Only follow these instructions to complete the calibration. When calibrating your machine the measured value of each parameter may not reflect the target values indicated on the sheet provided with the calibrator vial. Your goal is not to recreate the values on the target sheet provided with the calibrator vial; the targets are used as reference for calibrating the instrument. If you are in the middle of doing a calibration and need to call technical support make sure you return your calibration fluid to the refrigerator. 1. Press the QC Button 2. Press the Cal. Tab at the top of the screen 3. Press the Calibration Creation Button 18

Remove your calibrator vial from the refrigerator and follow the Blood Preparation instructions found in the Daily Quality Control instructions 6. Press the OK Button the calibration cycle will begin. 7. The needle will come down. Aspirate the sample by placing the vial under the needle and press the sample bar. 8. Results will be displayed. Press the OK Button to accept the results. 9. You can press the Cancel Button to discard a run and re-run the sample if you feel that the results are not satisfactory. If during any Run, the results are displayed and there is no OK Button, the machine won t accept that run due to a flag on one of the parameters. Press the Cancel Button to proceed. You will have to re-run the sample. If your calibration fails, return your calibrator vial to the refrigerator immediately and perform a concentrated cleaning procedure. Keep your print out and all related information and call technical support line for further instructions if your calibration continues to fail. You may be asked to fax your print out to the technical support fax line. 19
