2 Data Capture/Network Software Introduction The LS 6000 Data Capture/Network Software is a Microsoft Windows application program for IBM compatible personal computers and is provided as a 32-bit Version for Windows 95/98, and Windows NT 4.0 or later The software captures data from LS 6000 Series Scintillation Counter and stores them on a local drive on the PC or to a network drive, without user intervention. After the user has specified the file storage base directory and communication port parameters, the LS Data Capture/Network Software can be activated enabling it to capture data from the scintillation counter. Data Capture is able to run unattended in the background. Data received by the computer is stored in the selected base directory beneath user directories, similar to the directories created by the optional LS 6000 Data Buffer & Transfer System. Both the Data Buffer & Transfer System from Beckman and the LS 6000 Data Capture/Network Software store data in binary format called Beckman Standard File (BSF). This format provides certain benefits, such as: Compressed data; requires less disk space to archive data. Data cannot be modified, accidentally or on purpose (GLP/GMP). Each parameter has a unique code and contains the complete information. Specific data can be easily and accurately filtered out. Compatibility with Beckman applications software. However, binary files cannot be easily viewed and printed. Therefore, BSF may not be the desired format. The user may specify file conversions to ASCII Text and/or Lotus spreadsheet format in addition to the storage of BSF files. These file conversions will be performed automatically by Data Capture after a complete batch of samples has been received. This manual explains the use of the LS 6000 Data Capture/Network Software. Please refer to the LS 6000 or LS 6500 operating instructions from Beckman for information on the operation of the scintillation counter. 3 Getting Started Data Capture/Network Software Getting Started Make sure that the computer and the LS 6000 Scintillation Counter are properly connected via the serial communication port (RS-232 interface). Configure the program before capturing data from the scintillation counter to guarantee flawless data transfer. Select the Tools menu and then Options to open the configuration tabs and perform the following steps before activating the application: Select the Communication Port tab and set the communication parameters to match those defined in the LS 6000 Series Scintillation counter. Definethe Base Directory under the Directories tab. Select the desired File Formats (generic or per user) under the Directories tab. If desired, change any other default settings and click on OK. Activate the LS Data Capture/Network Software by clicking the Activate button on the Main Window or by selecting the Activate Data Capture item in the Capture menu. The LS Data Capture/Network Software is now ready to receive data. Note Make sure that the communication port settings on the LS 6000 Scintillation Counter match the settings in this program. Page-2
4 Data Capture/Network Software Getting Started Capturing Data To capture data from an LS 6000 Series Scintillation Counter, the program must first be activated. In the active state, the selected communication port is initiated and data sent from the scintillation counter are captured and saved on the selected local drive (hard disk) or on a network (remote) drive. If Data Capture is not active, the status bar displays the message Activate Data Capture. To prepare the application to capture data, press the Activate button or select the Activate Data Capture item in the Capture menu. After the application has been activated, the status bar will indicate Ready to receive data. The serial port light turns green, the port line lights CD (Carrier Detect) and DSR (Data Set Ready) turn red, and the arrow on the LS 6000 icon flashes in the colors red and yellow. The Capture menu item then changes to Cancel Data Capture and the Activate button changes to Cancel. Click on the Cancel Data Capture menu item or the Cancel button to deactivate the LS Data Capture/Network Software. Page-3
5 Getting Started Data Capture/Network Software Once the application has been activated, the program is able to run unattended in the background. The user may minimize the window by clicking the Minimize button or the Minimize Window item in the Capture menu. Data received from the scintillation counter are saved in BSF files that are named automatically following the specified filename convention. See Filename Conventions for details on this subject. Data Capture must be deactivated first to change any settings. To deactivate the application, click on the Cancel button or select the Cancel Data Capture item in the Capture menu. Data Capture will not receive data until it has been reactivated. If the computer running the LS Data Capture/Network Software was shut down, the application must be started and activated again. Up to four LS 6000 Scintillation Counters can be connected to a single computer for simultaneous data capture. Each LS 6000 connected to a common computer requires a unique installation of the LS Data Capture/Network Software in a separate directory on the computer. Page-4
6 Data Capture/Network Software Program Setup Program Setup Program Parameters To set up program and file conversion parameters, click on Options in the Tools menu and select the Program Parameters tab folder. Log Major Events Check the Log Major Events box if you wish to record capture, conversion and error events in a file. For more information on this function, see Logging of Events. Sound Support Check the Sound Support box if you want the program to use audible signals for major functions and errors. If a sound card is installed on your system, wave files will be played. Otherwise, the computer's beep function will be used. Include Error Messages in BSF File Check the Include Error Messages in BSF File box if you want the program to include error messages in the BSF file. Be aware that error messages in the BSF file can cause problems when using this file with applications programs. Include Counter ID in BSF File This function is only enabled when the user has entered a counter identification (serial number of the instrument or any other information) in the Counter ID text field. Check the Include Counter ID in BSF File box if you want the program to include the counter identification in the BSF file for GMP/GLP purposes. Page-5
7 Program Setup Data Capture/Network Software LS 6000 Series Counter The LS 6000 Series Scintillation Counter typically comes with 20 User Programs, which is reflected by the default setting. However, additional 30 User Programs are available to allow for a maximum of 50 User Programs. Click on the appropriate radio button to configure the program and match the configuration of your LS 6000 Series Scintillation Counter. Enter the serial number (S/N) of the instruments in the text field. Filename Convention The LS 6000 Data Capture/Network Software introduces a new scheme for automatically naming files. The default setting for Windows Data Capture reflects this. Those users that prefer previously used filename conventions may select either Similar to Data Buffer and Transfer System or Similar to DOS Data Capture. See Filename Conventions for details on this subject. Page-6
8 Data Capture/Network Software Program Setup Conversions File Formats The user may specify file conversions to ASCII Text or Lotus Spreadsheet format as well as applications software specific BSF formats. The file conversions will be performed automatically by Data Capture after a complete batch of samples has been received. The user may select one or more of the following file formats: Beckman Standard File (BSF) ASCII Text (ASC) Lotus Spreadsheet (WK1) ImmunoFit EIA/RIA (BSF) AccuFit (BSF) IsoMap 1 and 2 (BSF) The default format is Beckman Standard Format (BSF). Specific file format combinations are not allowed and the program disables the respective check boxes automatically. If the Individual User Formats box is not checked, the current selection applies to all users. For more information on this subject, see File Conversions. To examine examples of ASCII and Lotus files, see File Formats. Notes Lotus Spreadsheet (WK1) files have been originally defined by Lotus Development. The LS Data Capture/Network Software creates Version 2.0 Lotus files. Page-7
9 Program Setup Data Capture/Network Software Individual User Formats The default for all users is Beckman Standard Format (BSF). Check the Individual User Formats box to create user-specific file conversion profiles. In this case, a list box appears that shows the current file format selection for each user. To select user-specific file formats, simply scroll up or down to the find the appropriate user and click on it. The entry is highlighted and the File Formats frame allows you to make the desired file format selection for this user. A user that contains the entry >>> IPT <<< cannot be selected. This user number has been defined as the user program for Instrument Performance Tracking (IPT); a feature that will be implemented with version 2.0 of the software. Directories Base Directory The file storage location consists of: ABase Directory (full path, including drive and subdirectory) This can be a directory on the local drive (hard disk) or on a network (remote) drive. Removable drives, CD-ROM s, and RAM-Disks cannot be selected as file storage locations. The user subdirectory (USER 01 to 20 or USER 01 to 50) will be placed beneath the base directory that was created during setup. The user may separate files by type. In this case, file type subdirectories will be created automatically beneath the respective user subdirectory (BSF, ASCII, Excel and Lotus). The default base directory is a subdirectory of the program directory (<program path>\data). You may change the base directory and adapt it to your individual needs before you start capturing files. 10 Data Capture/Network Software Program Setup To change the base directory, click on Options in the Tools menu and select the Directories tab folder. In the Base Directory field, you may enter the full path for a new base directory or browse for an existing base directory by clicking on the button to the right of the field. Select the Browse button to the right of the base directory field to open the Browse dialog box and select an existing directory as the base directory. Select the desired drive and directory (double-click required) and choose OK. The selected drive and full path name will be transferred from the Browse dialog box to the Base Directory field. The program checks if the entry or directory selection is valid and warns the user, if necessary. The user must select a base directory on the local drive (hard disk) or on a network (remote) drive. Removable drives, CD-ROM's, and RAM-Disks cannot be selected as file storage locations. If the base directory is on a network drive, Data Capture must have the following access rights for the specified network drive and/or base directory: Generic access rights (drive must be mapped) Rights to create subdirectories, at least beneath the base directory Rights to create and delete files in the subdirectories Although the program checks the validity of the selected base directory in a generic fashion, we strongly recommend using the more comprehensive Network Access Test by clicking on the Test button. See Test Network Access for more information. File Format Directories Check the File Format Directories (BSF, ASCII, WK1) box if you desire to keep the various file types in separate directories. The directories will be created automatically and are located beneath the respective user subdirectories. The directory name for BSF files is BSF, the name for ASCII (ASC) files is ASCII and the name for Lotus Spreadsheet (WK1) files is LOTUS.Iftheboxisunchecked,allfileswillbestoredin the user directory. 11 Program Setup Data Capture/Network Software Communication Port Data Capture uses a serial communication port (RS-232 interface) to receive data from the scintillation counter. The current communication port settings are displayed in the status panels on the bottom left side of the main window. To change these settings, click on Options in the Tools menu and select the Communication Port tab folder. The user may change the Communication Port, Baud Rate, Parity, Stop Bits, and Data Bits as well as the Input Buffer Size. The Comm Port choices are COM1 to COM4. Unavailable ports are grayed-out and cannot be selected. The first available Communication Port (1 through 4) will be selected automatically by the LS Data Capture/Network Software, if it has not been set previously by the user. All baud rates supported by LS 6000 Series Scintillation Counters are supported by the LS Data Capture/Network Software. The Baud Rate, Parity, Data Bits, andstop Bits settings must match the settings on the scintillation counter. The default Input Buffer Size is 8K. With slow computers and when capturing linear spectrum data with a high baud rate, it might be necessary to increase the buffer size to 16K. The LS 6000 Series Scintillation Counter and the LS Data Capture/Network Software use XON/XOFF flow control. Page-10
12 Data Capture/Network Software Program Setup Test Network Access The Test Access button in the Directories tab is used to determine if Data Capture has the required access rights to the target drive and base directory. It should be run after a new base directory on a network (remote) drive has been selected. The Directory Access Tests will verify that Data Capture has three levels of access rights: 1. Generic access rights to the network drive and base directory 2. Rights to create subdirectories beneath the base directory 3. Rights to create and delete files in the subdirectories All three tests must pass to ensure the required access. If any test fails, the remaining tests will not be executed. In this event, see your Network Systems Administrator to make sure the network drive is mapped and that you have the required access rights to the specified target drive and base directory. Page-11
13 Program Setup Data Capture/Network Software Logging of Events The user may automatically record major events performed by the program as well as error conditions. Events include received files, converted files, as well as warning and fatal errors. All events are recorded with a date/time stamp. To record events, click on the Options item in the Tools menu, select the Program Parameters tab and check the Log Events box. Recorded events are kept in the file LSDC.LOG. The user may view and print the recorded events. File Name and Location Recorded events are kept in the file LSDC.LOG, which is located in the program directory shown in the top status bar. File Size The user may change the default size (8K) of the log file by dropping down the Max. File Size list box and clicking on the desired size. The choices are 4K, 8K, 16K, and 32K. Note The log file never exceeds the specified maximum file size. Old events are removed automatically so the file stays manageable and within the defined limit. Page-12
14 Data Capture/Network Software Program Setup Print Events Recorded events may be printed by clicking on the Print button. A window appears, allowing the user to specify printing of all or only selected pages. This button is disabled when the Log Events file has no entries. Printer Setup The Print Setup button opens a window that allows the user to select a printer as well as the print orientation (portrait or landscape). The currently selected default printer is shown and you may change the default printer from within this window. Refer to your Windows Operating Instructions for more information on printer setup functions. Purge All Events The Purge All button allows the user to delete all recorded events in the current file. A warning message appears when this button is clicked, giving the user the opportunity to abort the action. Page-13
15 LS 6000 Series Counter Data Capture/Network Software LS 6000 Series Counter Physical Connection A straight-through cable is required to connect the serial port of the computer with the serial port of LS 6000 Series Scintillation Counters. A cable with two 25-pin connectors and a 9-pin adapter is provided with the LS 6000 Data Capture/Network Software. Hardware and Software Requirements The LS 6000 Scintillation Counter requires an RS-232 interface. There are no other specific requirements to use LS 6000 Data Capture/Network Software with the counter. The LS 6000 Scintillation Counter allows the user to edit the type of information output through the RS-232 port. If the option to edit Output Formats is used, the following parameters for RS-232: Edit must be turned on for Data Capture to work properly: 1. User Number 2. Sample Number 3. Program Summary 4. CPM Average/DPM Average 5. Coefficient of Variation Communication Port Setup See Operating Instructions for Beckman LS 6000 or LS 6500 Scintillation Counter. Data Buffer & Transfer System The optional Data Buffer & Transfer System for LS 6000 Series Scintillation Counters (e.g., LS 6500, LS 6000SC, IC, TA, and LL) automatically creates BSF, ASCII, and Lotus files on diskette. The system stores the data in Beckman Standard File (BSF) format in the internal buffer and then converts them to ASCII and/or Lotus format, if requested. The LS 6000 Data Capture/Network Software does not necessarily replace the Data Buffer & Transfer System. To capture data with the LS 6000 Data Capture/Network Software, a computer connected to the counter is required. However, the LS 6000 Data Capture/Network software quickly and automatically saves data onto a hard disk or a network drive, without the memory limitations of a 1.44MB floppy disk. Benefits of LS 6000 Data Capture/Network Software BSF Files There are virtually no differences between BSF files created by the Data Buffer & Transfer System and the LS 6000 Data Capture/Network Software. However, the LS 6000 Data Capture/Network Software allows you to exclude error messages which can Page-14

It also provides the means to include the serial number of the counter in the BSF file. Although, BSF files are rather small compared to column-oriented ASCII and Lotus files, moving them from the data buffer to the floppy disk still requires some time. With the LS 6000 Data Capture/Network Software, files are available instantaneously. ASCII Files (ASC) ASCII files are text files and the user may select a column-oriented or delimiter format on both systems. Column-oriented ASCII files tend to become rather large and may take considerable time to move them from the Data Buffer to a floppy disk. ASCII files created by the Data Buffer & Transfer System do not contain column labels and the user may require the LS 6000 printout to determine which data are represented in the columns. Blank, %Reference Standard, Sample Repeat, and Sample Replicate results are suppressed and are not included in the ASCII file. Another limitation of the Data Buffer & Transfer System is that Sample Id strings are converted to numbers with the result that identifications such as B for Blanks, R for %Reference Standards, and S for Standards are eliminated. For a more convenient text format and to obtain an ASCII file with complete information, the LS 6000 Data Capture/Network Software is recommended. Also, the files are instantaneously available on your hard disk or a network drive without the space limitations of a 1.44MB floppy disk. Lotus Files (WK1) Lotus files created by the Data Buffer & Transfer System are virtually identical with those produced by the LS 6000 Data Capture/Network Software. The files become rather large and may take considerable time to move them from the Data Buffer to a floppy disk. With the LS 6000 Data Capture/Network Software, the files are instantaneously available on your hard disk or a network drive without the space limitations of a 1.44MB floppy disk. Page-15
17 Filename Conventions Data Capture/Network Software Appendix A Filename Conventions By default, the program uses the LS 6000 Data Capture/Network Software filename conventions, which provide the most complete and convenient information of the available choices. Other filename conventions are available for those users who wish to continue to use the filename conventions of the Data Buffer & Transfer System or the earlier DOS Data Capture product. However, these filename conventions are not as informative as the LS 6000 Data Capture/Network Software filename conventions. Please notice that the LS 6000 Data Capture/Network Software filename conventions do not include the user number within the filename. This is not required because files are always stored in user-specific subdirectories. When multiple CPM or DPM data sets are present in the original BSF file, two files must be created; one with CPM data from channel 1 or DPM results from isotope 1 and another with CPM data from channel 2 or DPM results from isotope 2. I BSF The first character identifies the application: (I = ImmunoFit, A = AccuFit and M = IsoMAP). The second character is a number and identifies the isotope or channel number. In this case, the original BSF file contained single CPM or DPM data. Positions 3 to 8 are described in Standard BSF Files (Data) above. Multiple Channels or Isotopes: When multiple CPM or DPM data sets are present in the original BSF file, two new files must be created; one with CPM data from channel 1 or DPM results from isotope 1 and another with CPM data from channel 2 or DPM results from isotope 2. I BSF The character 1 in Position 2 identifies the file containing the results from channel 1 or isotope 1. 21 Application BSF File Formats Data Capture/Network Software Appendix B Application BSF File Formats ImmunoFit EIA/RIA BSF files contain a reduced set of data. ImmunoFit EIA/RIA from Beckman requires Sample Id and CPM or Single-Label DPM for its radio immunoassay data reduction. Unnecessary data are eliminated to guarantee that the file is compatible with the applications program (see ImmunoFit & IsoMAP BSF Format, Dual-Label). AccuFit Fit Saturation and Competition BSF files contain a reduced set of data. Both AccuFit programs from Beckman require Sample Id, Rack Position, and CPM or Single-Label DPM for its receptor assay data reduction. Unnecessary data are eliminated to guarantee that the file is compatible with the applications program (see AccuFit BSF Format, CPM). IsoMap 1 BSF files contain a reduced set of data. IsoMap 1 from Beckman requires Sample Id and CPM or Single-Label DPM for its fraction analysis. Unnecessary data are eliminated to guarantee that the file is compatible with the applications program (see ImmunoFit & IsoMAP BSF Format, Dual-Label). IsoMap 2 BSF files are regular spectrum files. Multi-Label Data With data from more than one channel (e.g.; Dual- or Triple-Label DPM), the application conversion creates two files, one with CPM data from channel 1 or DPM results for isotope 1 and another with CPM data from channel 2 or DPM results for isotope 2. Data from channel 3 or isotope 3 are ignored. The filename indicates the application type as well as the channel or isotope (see Filename Conventions). If both CPM and DPM results are included in the original BSF file, DPM results are used and the CPM data are ignored. Note The header in application files is reduced to contain only essential file information to guarantee compatibility with the applications program. CPM data from channel 3 and DPM results for isotope 3 are ignored. Original BSF files which contain spectrum data, Single Rack Count data, Single Photon Monitor data or results from the Alpha/Beta Discrimination mode cannot be converted to an applications BSF file. 22 Data Capture/Network Software Alpha/Beta Discrimination Mode Appendix C Alpha/Beta Discrimination Mode When the Alpha/Beta Discrimination mode is selected in the user program, the BSF file contains information for a primary and a secondary isotope. The primary isotope can be defined in the user program; the default is Alpha. In the converted files, parameters for the secondary isotope in the header are marked with an uppercase B in front of the description. The column-oriented ASCII file always shows a for the primary isotope result and b for the secondary isotope. We recommend the use of the default settings in the scintillation counter for the primary isotope, so that a defines the Alpha and b the Beta emitter(s). Page-21
