The Zyla 5.5, five MP, 16 bit scmos USB camera system is used for spinning disc confocal and wide field fluorescence scanning (using the spinning disc retracted mode). The optional top of the line Hamamatsu Orca FLASH 4.0 V2 scmos cameras provides high speed multi-fluorescence scanning at full 4K resolution. On top of that, the camera has high quantum efficiency (82 % peak QE) and an excellent high signal to noise ratio as well as high contrast. Finally, the Pixelink M4 colour camera provides very fast brightfield scanning (TissueFAXS PLUS configurations). Fluorescence filter cubes are housed both in the spinning disc device (8 positions) and in the systems microscope main filter wheel (10 positions). 4
5 Zyla 5.5 Orca FLASH Pixelink M4 By using one or more multiband filter cubes in one or more of the filter turrets positions the high switching speed of the Spectra X light engine can be fully capitalized on as the filter turret will not have to change filter positions (or less often in case several multibands are used). Lightning Bolt Light Sources Brightfield light is provided by a VIS-LED with hours of life. Spectra X with comparison of its output intensity with Metal Halide and Xenon light sources VIS LED A Spectra X light engine is used for fluorescence illumination. This illumination system combines very high output, necessary for confocality, with very short switching times (< 10 ns rise and fall times between bands). The Spectra X uses six wide band solid state light sources with a fixed configuration, covering most of the spectrum. The user can change the emission filter paddles for the emission bandwidth needed. 5
