2 The fllwing sectins describe knwn issues with the Attune NxT v2.1 sftware. Where pssible we have utlined steps t wrk-arund the knwn issue. Guidance fr Instrument Start Up and Perfrmance Test using the Attune NxT v2.1 Sftware: Run perfrmance test directly after launching the sftware applicatin. If yu pen an experiment and then run perfrmance test yu may experience a failure. Restart the sftware applicatin and repeat perfrmance test if this ccurs. Knwn issue ( Artifact ) ID Number artf55035 artf55282 artf56497 artf56687 artf57858 Descriptin f Knwn Issue The Area scaling factr (ASF) is nt updated autmatically after baseline and perfrmance test are cmpleted. D nt press stp buttn during baseline as this can cause an intermittent timeut failure f subsequent baseline runs The Run and Recrd buttns may nt replace the Start Up buttn n the Cllectin panel fllwing instrument startup. This des nt happen with 100% frequency. When an instrument has an aut sampler attached, the sftware may smetimes becme unrespnsive if the user stps the Start Up functins after it has been started. Sftware can becme unrespnsive if start up is run prir t making sure all fluid bttles are plugged in Suggested Actin The Area scaling factr may be manually adjusted under the Advanced settings sectin f the Instrument Settings menu prir t acquiring and recrding a sample. If the area parameter lks larger than the height parameter, enter a value <1; if area lks smaller than height, enter a value >1. If a baseline is stpped and the next baseline run results in an errr dialg and r a baseline with everything failing, restart the applicatin sftware and the issue shuld be reslved It is recmmended that the user rerun Start Up and the buttns shuld becme active. DO NOT stp the Start Up functin after it has started. Pwer cycle the system with the fluid bttles plugged in Guidance fr Setting up an experiment using the Attune NxT v2.1 Sftware: Knwn issue ( Artifact ) ID Number artf53358 artf53507 Descriptin f Knwn Issue When adjusting the size f the plygn regin n a plt using the bunding rectangle bx, the regin may nt maintain their psitins. If the experiment name, grup, sample and parent gate are all displayed in a plt header, Suggested Actin Plygn gates can be made smaller r larger by mving the individual pints n the gate itself r by adjusting the bunding bx and then adjusting the individual pint that was nt maintained. When the plt type is changed using the rightclick custmize menu, the newly created plt will have the default scale f the previus plt. A plate experiment isn t autmatically active after it is created. The plate experiment is visible in the experiment explrer but is nt the experiment viewed in the Attune desktp. When creating experiments, Attune desn't check t see if it has enugh disk space t create the necessary files. This results in missing files and XML lad failures. If there isn't enugh rm t create the EE ndes, Errr -1 is returned, and attempting t pen any experiment that desn't cntain XML files, returns an undefined errr After an experiment is created the type f plate (96 r 384-well, as well as well type), cannt be changed using the custmize menu. shw the hyperlink. Decrease the length f the names t view the hyperlink r set the parent ppulatin by right-click within the plt t bring up a cntext menu where parent ppulatin may be selected. On custmize menu, deselect experiment and r sample name t further shrten the plt title label. If yu change plt types using the custmize menu, ensure that x-and y-axis scale ranges are set apprpriately. Duble click n the new experiment t activate it, create a wrkspace, run prtcl, and acquire samples. Archive yur data frm yur experiment explrer t free up disk space Ensure that the crrect plate type is selected upn experiment set up. If the wrng plate type is selected, delete the experiment and re-start. Guidance fr using the Experiment Explrer and Instrument Settings panel within the Attune NxT v2.1 Sftware: Target and Label Fields are applied t plt axes but d nt shw up in statistics tables By default, plt axes titles include the $PnS, r stain name, in additin t the target and label name. T remve the stain name and nly display the target and label name, select the plt and then select the custmize panel and delete the name in the axis label text bxes. When an FCS file is imprted t a sample, the instrument settings panel des nt update t the sample specific data. It will display the experiment level instrument settings. The target and label names and parameter list are crrect in the drpdwn list n a wrkspace plt. Guidance fr using Cmpensatin within the Attune NxT v2.1 Sftware: If yu make an errr in the cmpensatin set up wizard and need t make changes, yu must delete and then re-start the cmpensatin set up prcess. The gate names n histgram plts within cmpensatin samples are shwn in large text and cannt be custmized If cmpensatin fr the experiment has never been set up and yu imprt an FCS file with embedded cmpensatin values, the cmpensatin values will nt be cmpensated and yu will need t push the Apply Cmpensatin buttn. The HyperLg scale is nt available fr cmpensatin cntrl samples using the right click menu. If a sample has an FCS file with an embedded spillver matrix the matrix frm the FCS file will nt be able t be used after the file is imprted int a new experiment. The Cmpensatin Setup buttn may be available frm the cmpensatin ribbn during acquisitin Unding the mvement f a gate n a cmpensatin cntrl will nt update the cmpensatin matrix Suggested Actin Ensure that the desired area r height parameter is selected prir t selecting parameters fr cmpensatin cntrls. If area r height measurement is changed, ensure that the crrect parameters are selected prir t sample acquisitin. T set the axis f cmpensatin cntrls using a HyperLg axis scale, adjust the axis scale using the custmize panel. If an FCS file is imprted int an experiment, enter spillver values int the spillver matrix manually by selecting the View Matrix buttn f the Cmpensatin tab f the Ribbn bar. Ensure that OK r enter are pressed fllwing data entry. D nt launch cmpensatin setup during acquisitin. It is best practice t set up cmpensatin prir t starting yur experiment t ensure crrect instrument set-up. D nt use the und functin when making adjustments t the gates r markers n the cmpensatin wrkspace. Adjust the gates back t the previus psitin if necessary Rev. 5 Guidance fr Acquiring Samples using the Attune NxT v2.1 Sftware: The default setting fr FSC vltage is set very lw; increase t at least 400 t put mst cmmn cell types n scale using a FSC vs. SSC scatter plt. At very high event rates n very large data cllectins, yu may bserve a slwing f the data update n the screen (~5 sec between updates). This is expected behavir and des nt have any impact n the cmpletin f the acquisitin. Keep the tube lifter in the DOWN psitin when using the aut sampler. Derived gates cannt be used as a stp ptin. Knwn issue ( Artifact ) ID Number artf56764 artf57941 Descriptin f Knwn Issue Lad plate int aut sampler dialgue bx will be shwn if the Attune Aut Sampler is unplugged while the sftware is pen When adding mre sample r trying t acquire mre events, the cmplete stp cnditin ptin is nly available fr the first additin t time, vlume, and events stp criteria. After that, yu will nly be able t append data nt cmplete the stp criteria therefre amunt left f stp criteria will nt apply anymre. Suggested Actin D nt unplug the aut sampler while the sftware is pen. If the lad plate int aut sampler dialgue is bserved after unplugging the system, click the cancel buttn t remve the dialgue bx. Only use the cmplete stp cnditins ptin nce. Guidance fr using the Experiment Wrkspace (Gates, Plts, Stats) within the Attune NxT v2.1 Sftware: With large event files, there can be a slw respnse in between cmmands. Faster respnse rates can be achieved with fewer parameters selected and using a decreased number f plts and gates n wrkspace. The time it takes t pen the plt Preview panel is dependent upn the number f parameters enabled Suggested Actin Keep plt reslutin greater than 256x256 fr best visualizatin. Reslutin can be adjusted under the Custmize panel when a plt is selected. Names and labels can be truncated depending upn the fnt size used and r length f name. Changing the crdinates r rtatin f an val gate using the custmize menu des nt wrk. The icns n the wrkspace tlbar are nt active when a new plate nde r experiment is activated Plt titles in aut layut mde are limited t 35 characters and the title including Experiment, Grup, Sample, and Parent Gate will nt be visible when >35 characters are used in aut layut. T crrect this press the Esc buttn n the keybard. Resize and reshape an val gate thrugh manipulatin f the bject within the plt. Define a grup/sample and activate a well sample fr the wrkspace tl bar t becme active Keep plt titles t less than 35 characters by limiting title name r d nt include hierarchical elements (such as Experiment, Grup and Sample) in title name. Alternatively view the plt in freefrm mde and resize the plt until all elements are visible. Guidance fr Data Analysis / Data Display using the Attune NxT v2.1 Sftware: On the Results view, right clicking n a clumn header will display a list f statistics, sme f which are nt currently available. The Results Table can be cpied but the header rw is NOT cpied and must be entered int a secndary data analysis prgram such as Micrsft Office Excel manually. Knwn issue ( Artifact ) ID Number artf55357 artf56533 Descriptin f Knwn Issue The results table will revert t default settings and nt maintain user-selected statistics nce the sftware is clsed. A statistics bx will reprt a statistical value fr a disabled parameter if the experiment wrkspace is made that includes a parameter that is disabled AFTER the wrkspace is finalized. In this instance the statistics bx inserted int the wrkspace will shw a value f 0 fr every measure assciated with the disabled parameter instead f ignring the disabled parameter. Suggested Actin The statistical measure f interest can be recalculated by the sftware by selectin f the measure frm the Statistics ribbn. Re-select the statistics f interest when the sftware is re-launched. D nt include plts in a wrkspace fr disabled parameters. If a plt is inserted in a wrkspace fr a disabled parameter, a brick symbl will appear n the plt. Delete this plt befre inserting the statistics bx Rev. 7 Guidance fr Exprt Imprt f Experiments, FCS files using the Attune NxT v2.1 Sftware: The sftware supprts FCS3.1 frmats Knwn issue ( Artifact ) ID Number artf56354 artf57789 Descriptin f Knwn Issue The first recrded FCS file in an experiment is recrded as DefaultSampleName.fcs, even if the name is changed. All subsequent fcs files will be recrded with the crrect file name. Target and label name edits made after recrding a sample are nt updated in FCS file. Suggested Actin If using a 3 rd party analysis sftware package, make all changes t target and label fields prir t recrding. 8 Installatin Instructins: System Requirements: Quad cre prcessr, 16 GB RAM, 500 GB disk space available. Windws 7 64 bit Prfessinal with Service Pack 1. Attune Sftware v2.1 Installatin NEW INSTALL (If upgrading frm a previus versin, see sectins belw) Sftware Installatin Nte: D nt launch the Attune NxT applicatin until the cmpletin f all the installatin steps. 1. Restart the cmputer befre beginning these instructins. 2. Lg int Windws as an administratr. (Unless changed the default administratr user accunt is: INSTR-ADMIN and passwrd: INSTR-ADMIN). Nte: It is recmmended that n ther applicatins are running while cmpleting these steps. 3. Unzip (select extract all files ) the AttuneNxTv2.1.zip 4. Open the USB3_FOR NEW INSTALL, INSTALL ME FIRST flder and duble-click the RENESAS- USB3-Hst-Driver setup.exe file. 5. Fllw the instructins and accept the terms f agreement. Once cmplete, click Finish. 6. Plug in the Deskey device and pwer n the Attune NxT cytmeter 7. Duble-click setup.exe in the Attune NxT_Build flder. 8. During the installatin prcess described belw, yu may have a windws ntificatin ppup frm the tlbar stating that a driver installatin was nt cmpleted. These messages can be ignred. Restart the cmputer and cytmeter befre beginning these instructins. 2. Lg int Windws as an administratr. (Unless changed the default administratr user accunt is: INSTR-ADMIN and passwrd: INSTR-ADMIN). Nte: It is recmmended that n ther applicatins are running while cmpleting these steps. 3.
