7 Chapter 1 Intrductin U.S. FDA 21 CFR Part 11 Cmpliance CFX Maestr sftware, Security Editin enables CFX Maestr users t meet the Fd and Drug Administratin s regulatins n gd labratry practices in the pharmaceutical and bitechnlgy industries. When enabled, Security Editin prvides the necessary features t permit CFX Maestr t perate in cmpliance with Title 21 f the U.S. Cde f Federal Regulatins Part 11 (21 CFR Part 11) within a clsed system. A clsed system is defined as an envirnment in which system access is cntrlled by the persns wh are respnsible fr the cntent f electrnic recrds that are n the system (Sectin 11.3 (b) (4)). Nte: The security cntrls built int CFX Maestr must be prperly cnfigured and administered by the CFX Maestr administratr(s) in yur rganizatin in rder t be secure and in cmpliance with 21 CFR Part 11. Bi-Rad makes n claim that CFX Maestr sftware, Security Editin is CFR cmpliant in and f itself, nr des the cmpany guarantee cmpliance fr the user. Yur rganizatin must establish plicies and standard perating prcedures that wrk in cnjunctin with the tls prvided by Bi-Rad t ensure cmpliance with 21 CFR Part 11. 9 Chapter 2 Activating CFX Maestr Sftware, Security Editin Imprtant: CFX Maestr sftware, Security Editin uses Micrsft Windws User Authenticatin t verify users access t the sftware. All users wh plan t access Security Editin must be able t lg in t the CFX Maestr cmputer with their Windws credentials. By default, CFX Maestr sftware, Security Editin (knwn in this dcument as Security Editin) starts in standard mde. Security mde is enabled when yu insert the Security Editin dngle int a USB prt n the CFX Maestr cmputer. CFX Maestr sftware remains in security mde until yu remve the dngle. As in standard mde, when security mde is enabled CFX Maestr detects the supprted thermal cyclers and displays them in the Detected Instruments pane. In security mde, the thermal cycler must be cnnected t the CFX Maestr cmputer by USB cable. The S1000 thermal cycler must be cnnected t a C1000 Tuch thermal cycler by USB cable. Imprtant: The MiniOpticn thermal cycler is supprted in security mde nly n cmputers running the Micrsft Windws 7 perating system. CFX Maestr sftware des nt recgnize the MiniOpticn thermal cycler n cmputers running the Micrsft Windws 10 perating system. User Guide 7

10 Chapter 2 Activating CFX Maestr Sftware, Security Editin Activating Security Editin Imprtant: Activating Security Editin requires yu t be a member f the Windws Administratrs grup n the CFX Maestr cmputer. Ensure yu meet this requirement befre yu begin. This prcedure assumes that yu have successfully installed the current versin f CFX Maestr sftware. See the dcument CFX Maestr Sftware User Guide fr mre infrmatin. T activate Security Editin 1. If yu have nt already dne s, install CFX Maestr sftware (knwn in this dcument as Standard Editin). 2. Cnnect a supprted thermal cycler t the CFX Maestr cmputer. 3. Start CFX Maestr sftware and verify that the attached thermal cyclers appear in the Detected Instruments panel. 4. Shut dwn CFX Maestr sftware. 5. Insert the supplied Security Editin dngle int a USB prt n the CFX Maestr cmputer and restart the applicatin. 6. Lg in t Security Editin with yur Windws credentials. The sftware verifies yur Windws accunt and starts in security mde. After activating Security Editin, yu must create a user accunt fr each user wh plans t access the sftware. 11 Managing Security Editin User Accunts Managing Security Editin User Accunts In Security Editin, users lg in t CFX Maestr sftware with their Windws user name and passwrd. The persn wh installed Security Editin is autmatically assigned the rle f CFX Maestr Administratr and can create and manage Security Editin user accunts and rles. All ther users must be assigned a Security Editin user accunt in rder t lg int and use the sftware. This sectin explains hw t manage Security Editin user accunts. Imprtant: Each user must have a Windws accunt and passwrd n the CFX Maestr cmputer befre yu can assign a Security Editin user accunt and rle. Users can be members f either the Windws Users grup r Windws Administratrs grup. Members f the Windws Users grup can access nly their wn files and flders (including Security Editin files and flders). Members f the Windws Administratrs grup can access the files and flders f ther users n the cmputer (including ther Security Editin files and flders). Review the Micrsft Windws Administratin dcumentatin and see yur Windws system administratr fr mre infrmatin befre creating Windws accunts. Managing Security Editin Users Tip: Only users with the CFX Maestr Administratr rle can create and remve Security Editin user accunts. The persn wh installed CFX Maestr sftware is autmatically assigned the CFX Maestr Administratr rle. That persn can assign the Administratr rle t ther users. Nte: In Security Editin, at least ne user must be assigned the Administratr rle. T add Security Editin user accunts 1. Verify that each Security Editin user is a member f either the Windws Users grup r Windws Administratrs grup and has a Windws passwrd n the CFX Maestr cmputer. Tip: Review yur Micrsft Windws Administratin dcumentatin r see yur Windws system administratr fr infrmatin abut creating Windws users and grups and setting Windws passwrds fr each user. 2. Start Security Editin and lg in as the sftware administratr. 3. In the Hme windw, select User > User Administratin. User Guide 9

In the Manage Users sectin, prvide the fllwing infrmatin fr each user: User name in Security Editin, this must be the user s Windws lgin user name. Full name the user s full name. This name appears in the Full User field in the audit trail. Rle the rle t assign t the user. Nte: Yu can select nly ne rle frm the drpdwn list. See Managing Security Editin User Rles n page 11 fr mre infrmatin. Dmain the Windws dmain frm which the user accesses the sftware. See yur Windws system administratr fr mre infrmatin. 5. 13 Managing Security Editin User Accunts T remve a Security Editin user accunt 1. Start Security Editin and lg in as a sftware administratr. 2. In the Hme windw, select User > User Administratin t pen the User Administratin dialg bx. 3. In the Manage Users pane, select Remve fr each user yu want t remve. 4. Click OK and then click Yes t save changes and clse the User Administratin dialg bx. Managing Security Editin User Rles Imprtant: Security Editin requires that at least ne user be assigned the Administratr rle. Yu can assign mre than ne user this rle. Security Editin has fur user rles. Each user must be assigned a rle in rder t access the sftware. Althugh users can be assigned nly ne rle, yu can change a user s rle at any time. The default rights fr rles in security mde are the same as thse in standard mde. Except fr the Administratr rle, yu can change the rights assigned t each rle. All users assigned a rle inherit nly the rights f that rle. By default, the rights fr each rle are as fllws: Administratr this rle has all rights; yu cannt change these rights. Principal this rle has all rights. Operatr this rle has all rights except skipping cycles. Guest this rle can nly read files. When assigning rles in security mde, carefully determine the requirements fr each user. Fr example, withut the right t save, users assigned the Guest rle will nt be able t sign a file. T mdify the rights fr a rle 1. Start Security Editin and lg in as a sftware administratr. 2. In the Hme windw, select User > User Administratin t pen the User Administratin dialg bx. 3. In the Manage Rights sectin, fr each rle clear r select the checkbx f specific rights as necessary. 4. Click OK and then click Yes t save changes and clse the User Administratin dialg bx. 14 Chapter 2 Activating CFX Maestr Sftware, Security Editin Starting Security Editin T start Security Editin, each user must lg in with their Windws user name and passwrd. T start Security Editin 1. Click the CFX Maestr shrtcut icn t start the applicatin. 2. In the Lgin dialg bx, type yur Windws passwrd and click OK. Security Editin pens t the Hme windw. The title bar displays the Windws user name f the lgged in user, and the menu bar displays a blue sticker indicating the sftware is 21 CFR Part 11 cmpliant, fr example: 12 CFX Maestr Sftware, Security Editin

15 Chapter 3 Using CFX Maestr Sftware, Security Editin When Security Editin is enabled, CFX Maestr sftware remains in security mde until yu remve the dngle. Imprtant: All files saved in Security Editin are marked as secure files and cannt be pened in standard editin. This chapter explains hw t use CFX Maestr sftware, Security Editin. Using Security Editin As in standard mde, when security mde is enabled CFX Maestr sftware detects the supprted thermal cyclers and displays them in the Detected Instruments pane. In security mde, the tuch screen displays ntificatin that the system is under remte cntrl, fr example: When the thermal cycler is under remte cntrl, users can d the fllwing: View a run in prgress. View the instrument clck. User Guide 13

16 Chapter 3 Using CFX Maestr Sftware, Security Editin View the status f the plate. View the curves fr the current run. Pause the run. Skip a step. Select wells n the plate. Cancel a run. Open the lid. When the thermal cycler is under remte cntrl, users cannt create plates r prtcls and all runs must be initiated frm the CFX Maestr cmputer t which the system is attached. The resulting.pcrd file that is saved directly nt the CFX Maestr cmputer is saved as a secure data file. It can be pened nly in security mde. Imprtant: The resulting.zpcr file is nt saved as a secure zip file. Users can pen the.zpcr file n a cmputer running CFX Maestr in standard mde and can save the.pcrd files as unsecure. Ensure that the cmputer is running CFX Maestr in security mde befre pening the.zpcr and saving the data files. T retrieve the secure data file 1. D ne f the fllwing: In the sftware, right-click the instrument in the Detected Instruments pane and select Retrieve Data File. On the thermal cycler, insert a USB drive int the USB prt and cpy the file t the drive. 2. Save the data file t a shared flder n the CFX Maestr cmputer r t yur persnal flder. Nte: Only Security Editin users with a Windws lgin and the apprpriate rle n the CFX Maestr cmputer can pen the secure data file. 14 CFX Maestr Sftware, Security Editin

17 Using Security Editin n the CFX Maestr Cmputer Using Security Editin n the CFX Maestr Cmputer Imprtant: Security Editin uses Micrsft Windws User Authenticatin t verify access t secure CFX data files. Cntact yur Windws administratr t create an envirnment that cmplies with 21 CFR Part 11 requirements. All features in CFX Maestr standard mde are available in security mde. In additin, in security mde users can d the fllwing: Sign data and gene study files. Passwrd prtect data files. View and print audit trails. This sectin explains these features in detail. Secure Files By default, CFX Maestr sftware saves secure files t the lgged in user s persnal Security Editin flder, which is lcated at c:\users\<username>\my Dcuments\Bi-Rad\CFX\My qpcr Security Editin sets this flder t read-nly. Hwever yu can share this flder and its cntents with thers n yur team. Tip: Alternatively, yur Windws system administratr can create a shared flder and yur Security Editin administratr can prgram the sftware t save all files t that flder. In Security Editin, plate, prtcl, data, and gene study files are marked as secure when they are saved. Yu can create these files in standard mde r in security mde. After they are saved in security mde, yu can pen these files nly in security mde. Security Editin creates an audit trail fr all secure data and gene study files (.pcrd and.mgxd files, respectively). The sftware recrds all auditable activity in the file s audit trail. Fr mre infrmatin, see Audit Trails n page 24. Signing Secure Files After a file is saved in security mde, users can add an electrnic signature. T sign a file, the user s rle must have the right (r permissin) t save a file. Fr example, by default the Guest rle des nt have the right t save a file and therefre users assigned this rle cannt sign a file. User Guide 15

18 Chapter 3 Using CFX Maestr Sftware, Security Editin In Security Editin, signed files are nt set t read-nly. They can be reviewed, mdified, and signed multiple times. All changes and signatures are tracked in the file s audit trail. Yu can sign the fllwing file types: Data files (.pcrd) Gene study files (.mgxd) Nte: Files must be saved in security mde befre they can be signed. If yu have recently perfrmed a run in security mde, save the resulting data file first. T sign a file 1. Lg in t Security Editin with yur Windws lgin credentials. 2. Open the secure data file r gene study file t sign. 3. Chse File > Sign. The Electrnic Signature dialg bx appears. 4. Enter yur Windws user name and passwrd and the reasn fr signing the file. The user name and reasn fr signing are included in the audit trail (fr mre infrmatin see Audit Trails n page 24). 5. 19 Using Security Editin n the CFX Maestr Cmputer Mdifying Secure Files In Security Editin, users can mdify secure files, including signed and unsigned data and gene study files. The sftware prmpts yu t prvide a reasn fr the change when yu save a mdified secure data r gene study file. The changes are tracked in the file s audit trail. Tip: Because the sftware des nt create audit trails fr plate r prtcl files, yu are nt prmpted t prvide a reasn when yu save changes t these files. T save a mdified data r gene study file 1. Lg in t Security Editin with yur Windws lgin credentials. 2. Open and mdify a secure data file r gene study file. Tip: Fr a list f auditable activities, see Auditable Events n page Chse File > Save. The Audit Trail Change Reasn dialg bx appears. This dialg bx displays the fllwing infrmatin, which is captured in the file s audit trail header fr each mdificatin event: Date the date n which the change ccurred. User the Windws dmain and user name f the lgged in user. User Guide 17

Applicatin CFX Maestr sftware (appears as BiRadCFXManager.exe, which is crrect). Applicatin versin the current versin f CFX Maestr sftware, Security Editin. Full user the full name f the lgged in user. Nte: This name appears in the audit trail. Machine the cmputer n which Security Editin is installed. The change table displays the auditable changes that ccurred as a result f the mdificatin. A brief descriptin fr the reasn f the change might als appear. Tip: Yu can add r edit descriptins in the Reasn fr Change clumn. 4. Review the list f changes. Prvide detailed reasns if necessary. 5. D ne f the fllwing: Click Save Changes t save the changes t the file as well as any changes yu made t the table and clse the dialg bx. The changes t the file and the reasns fr the changes appear in the file s audit trail. Click Cancel Changes t revert the file t its previus state and clse the dialg bx. 21 Using Security Editin n the CFX Maestr Cmputer Passwrd Prtecting Secure Files As an added level f security, Security Editin enables users t set passwrds n all secure files. When setting passwrds n a secure file, cnsider the fllwing cnditins: Cnditin N passwrd is required. File requires the Save passwrd. File requires the Open passwrd. File requires bth the Open and Save passwrds. Actin All users can pen, mdify, and save the secure file. All users can pen the secure file, and users wh knw the Save passwrd can mdify and save the secure file. Only users wh knw the Open passwrd can pen, mdify, and save the secure file. Sme users can pen the secure file, and a subgrup f thse users can mdify and save the file. Depending n the user s rle, any user can perfrm Save As t create a new secure file with anther name r save a file with the same name in anther lcatin as lng as ne f the fllwing is true: The secure file is nt passwrd prtected. The user has the passwrd t pen the file. Tip: The new file is saved withut passwrd prtectin. The riginal file retains its passwrds. Depending n the rle, a user can mdify and save the riginal file as lng as ne f the fllwing is true: The file is nt passwrd prtected. The user has the passwrd t pen and the passwrd t save the file. Nte: The user s rle must include the right t save files in rder t set passwrds. Fr example, users with the rle Guest cannt save files and therefre cannt set passwrds n a file. Imprtant: Only Security Editin administratrs can reset passwrds. User Guide 19

22 Chapter 3 Using CFX Maestr Sftware, Security Editin T passwrd prtect a file 1. Lg in t Security Editin with yur Windws credentials. 2. Open the secure file. 3. Chse File > File Passwrds. The File Passwrds dialg bx appears. 4. Enter passwrds in the Save Passwrd and Open Passwrd bxes. Tip: By default, passwrds appear as asterisk characters when typed. Select Shw Passwrd t display the passwrd as yu type it. Imprtant: Passwrds are case sensitive. Security Editin des nt set limitatins n passwrds. Fr best practice, see yur system administratr fr passwrd requirements at yur site. 5. Click Set Passwrds t set the passwrds and clse the dialg bx. 6. 23 Using Security Editin n the CFX Maestr Cmputer T remve passwrds 1. In the Passwrd Required dialg bx, click Remve Passwrds. Nte: Yu must be a Security Editin administratr t remve passwrds. The CFX Maestr Security Editin Lgin dialg bx appears. 2. Prvide the Windws user name and passwrd fr the Security Editin administratr and click OK. The riginal data file appears. Imprtant: Yu must save the file in rder t remve the passwrds. 3. Chse File > Save t save the changes t the file. User Guide 21

Tip: Save Passwrd, Open Passwrd, and Shw Passwrd are disabled. 3. Click Reset Passwrds. Nte: Only Security Editin administratrs can reset passwrds. 25 Using Security Editin n the CFX Maestr Cmputer 4. Prvide the Windws user name and passwrd fr the Security Editin administratr and click OK. The File Passwrds dialg bx appears. 5. D ne f the fllwing: T reset passwrd prtectin, type a new passwrd in the apprpriate passwrd bx. T remve passwrd prtectin, clear the passwrd bxes. 6. Click Set Passwrds t save the passwrd changes and exit the dialg bx. User Guide 23

26 Chapter 3 Using CFX Maestr Sftware, Security Editin Audit Trails Security Editin creates audit trails fr data and gene study files (.prcd and.mgxd files respectively). Any changes made t r actins perfrmed n secure data and gene study files are captured in the file s audit trail when the file is saved. Security Editin creates a separate audit trail fr each file. Yu can chse File > Save As and save secure signed r unsigned data and gene study files int anther flder r with anther name. The new file inherits the audit trail frm the riginal file. The audit trail fr the new file als includes the Save As activity. Changes t r actins perfrmed n the new file are captured in its wn audit trail. The riginal file retains its audit trail, in which further activity is captured. Auditable Events n page 26 lists the auditable events that the sftware captures. 27 Using Security Editin n the CFX Maestr Cmputer T view the audit trail u In the pen data r gene study file, select View > Audit Trail. The file's audit trail appears. By default, the data are srted by date and time, and all events appear in the expanded view. Yu can filter the view by user name and bject, and cllapse the expanded view t easily srt by any header field. Yu can als view the audit trail as an html reprt. T srt by user name u Select the target user frm the User Name drpdwn list. T srt by bject u Select the target frm the Object drpdwn list. T hide the full descriptin f events u Click Cllapse All. T srt data in the change detail table u Click the diamnd symbl in the data clumn header t perfrm an ascending srt (A t Z, smallest number t largest, r earliest t mst recent). T print the audit trail 1. Click HTML Reprt t display the audit trail in a web brwser. 2. In yur brwser windw, d ne f the fllwing: Select File > Print. Right-click the reprt and select Print.
